Scope

GB/T 21675-2022 is the English-translated version of 非洲马瘟诊断技术.

GB/T 21675-2022 is the Chinese national standard for diagnosing African horse sickness, a viral disease of equines caused by an orbivirus with nine known serotypes and spread by Culicoides midges.

It sets out the biosafety measures, the clinical diagnosis - susceptible animals, clinical signs, pathological changes and clinical judgement - the collection and handling of samples, and the laboratory methods: virus isolation and identification, RT-PCR, general and type-specific real-time RT-PCR, and indirect and blocking ELISA for antibodies, followed by the rules for the overall judgement.

Annexes give the solutions used and the primer and probe sequences. It applies to the diagnosis, detection, quarantine, surveillance and epidemiological investigation of the disease in horses, mules, donkeys and other equines, and cites GB 19489.

It replaces GB/T 21675-2008, was issued on 30 December 2022 and has applied since 1 July 2023.

Foreword

This document is in accordance with the provisions of GB/T 1.1-2020 "Guidelines for Standardization Work Part 1: Structure and Drafting Rules for Standardization Documents" drafting.

This document replaces GB/T 21675-2008 "African Horse Sickness Diagnostic Technique". Compared with GB/T 21675-2008, except for structural adjustment and Apart from editorial changes, the main technical changes are as follows.

--- Added the chapter "Abbreviations" (see Chapter 4);

--- Added "biosecurity measures" (see Chapter 5);

--- Added "General Real-timeRT-PCR method" (see 8.3);

--- Added "type-specific Real-timeRT-PCR method" (see 8.4);

--- Added "blocking ELISA antibody detection method" (see 8.6);

--- Deleted "separation of suckling mice" (see 4.1.4 of the 2008 edition);

--- Deleted "micro-complement fixation reaction" (see 4.2.2 of the 2008 edition).

Introduction

A non-contact viral infectious disease caused by equine animals, characterized by changes in the respiratory system and circulatory system, often causing horses, mules At least two species of Culicoides can spread the disease. AHSV belongs to the genus Orbivirus of the family Reoviridae. The disease is currently known to have 9 serotypes type. The World Organization for Animal Health (WOAH) lists it as a notifiable animal disease. my country defines it as a first-class animal disease.

The occurrence of this disease has a certain seasonality and regionality, and it is more common in hot and humid seasons. Once it is introduced, it is often endemic or outbreaks. broadcast quickly. Thick frost, high dry terrain, natural barriers and other climatic and geographical conditions that affect the reproduction or activities of vector insects can significantly reduce the disease.

The content of diagnostic techniques in this document includes clinical diagnosis, etiological diagnosis methods and serological diagnosis methods.

The revision of this document refers to the WOAH Manual of Diagnostic Tests and Vaccines for Terrestrial Animals (2019), and incorporates relevant Chinese technologies.

1 Scope
