3 Terms and definitions
The following terms and definitions apply to this standard.
3.1 Microbial inoculants in agriculture
The live bacteria preparations, which are processed from target microorganisms (effective bacteria), through industrialized production and propagation.
4 Product classification
According to the types or functional characteristics of the contained microorganisms, it can be divided into rhizobia inoculants, nitrogen-fixing bacteria inoculants, phosphate-dissolving microbial inoculants, silicate microbial inoculants, photosynthetic bacteria inoculants, decomposing agent for organic materials, growth-promoting bacteria, mycorrhizal inoculants, bioremediation inoculants, etc.
5 Requirements
5.1 Bacteria species
The microbial strains, which are used in production, shall be safe and effective.
The producer shall provide the classification and identification report of the bacterial species, including complete information such as the scientific name of the genera and species, morphology,
5.2 Product appearance (sensory)
Powder products shall be loose. Granular products shall have no obvious mechanical impurities, be uniform in size, have water absorption.
5.3 Product technical index
5.3.1 The technical index of agricultural microbial inoculants products is as shown in Table 1, wherein the technical index of organic material decomposing agents are implemented in accordance with Table 2.
5.3.2 See Table 3 for harmlessness index of the microbial inoculants in agriculture.
6 Test method
6.1 Instrument and equipment
6.1.1 Biological microscope
6.1.2 Constant temperature incubator
6.1.3 Constant temperature drying oven
6.1.4 Ultra-clean workbench or clean room
6.1.5 Electronic balance (or precision balance)
6.1.6 Shaker
6.1.7 Steam sterilization pot
6.1.8 Test sieve
6.1.9 Acidity meter
6.2 Reagents
6.3 Testing of product parameters
6.3.1 Determination of appearance (sensory)
Take a small amount of samples. Place them in a white enamel tray (or white plastic palette). Carefully observe the color, shape, texture of the sample.
6.3.2 Determination of effective viable bacteria count
Use the plate counting method. Select an appropriate medium, according to the type of microorganisms tested.
If the dilution method [Most Probable Number (MPN) 5 pipe method] is used, it is performed in accordance with the provisions of Appendix C.
6.3.2.4 Colony count
Take the dilution plate, which appears 20 ~ 300 colonies, as the counting criteria (10 ~ 150 colonies for filamentous fungi). Respectively count the number of effective viable bacteria and the number of miscellaneous bacteria. When there is only one dilution AND the average number of colonies is between 20 ~ 300, the average number of colonies is used for calculation.
6.3.3 Determination of the number of fungi
Use Martin medium. The determination method is the same as 6.3.2.
6.3.4 Determination of the rate of miscellaneous bacteria
Except for the effective bacteria in the sample, all other bacteria are miscellaneous bacteria. The rate of miscellaneous bacteria in the sample is calculated according to formula (3).
6.3.7.3 Particle samples
The sample is first crushed and pass through a 1.0 mm test sieve. Make measurement, according to the method of 6.3.7.2.
6.3.8 Determination of the number of fecal coliforms
It shall meet the requirements of GB/T 19524.1.
6.3.9 Determination of the mortality of ascaris eggs
It shall meet the requirements of GB/T 19524.2.
6.3.10 Determination of cellulase activity and protease activity
It shall meet the requirements of Appendix D.
6.3.11 Determination of arsenic, cadmium, lead, chromium, mercury
It shall meet the requirements of 5.12 ~ 5.17 in GB 18877-2002.
6.3.12 Inspection of shelf life
Before the shelf life indicated in the product manual, measure the corresponding indicators of the product, according to the methods 6.3.1 ~ 6.3.11.
7 Inspection rules
The digital rounding of product technical index in this standard shall comply with the provisions of GB 8170. The qualification judgment of product quality shall comply with the provisions of the comparison method of rounding values in GB 1250-1989.
7.1 Sampling
The products, which are processed from each fermenter's bacteria liquid (or each batch of solid fermentation), as a batch, to carry out sampling inspection.
During the sampling process, it shall strictly avoid the contamination of bacteria.
7.1.1 Sampling tools
Sterile plastic bags (bottles), metal spoons, samplers, measuring cylinders, kraft paper bags, glue, sampling seals, sampling sheets, etc.
7.1.2 Sampling method and quantity
Generally, the sampling is carried out in the finished product warehouse, through the random method.
7.2 Inspection classification
7.2.1 Exit-factory inspection
7.2.1.2 The exit-factory inspection items are the items specified in 5.2 and 5.3.1, but do not include the shelf life.
