Scope

GB/T 35808-2018 is the English-translated version of 林业生物质原料分析方法 纤维素酶活性测定.

GB/T 35808-2018 is the Chinese national method for measuring cellulase activity when forestry biomass is analysed. The enzyme hydrolyses sodium carboxymethyl cellulose at a set temperature and pH, the reducing sugars released are made to react with 3,5-dinitrosalicylic acid (DNS), and the colour formed is read at 540 nm against a standard curve.

The standard gives the terms and definitions, including the cellulase activity unit, the principle, the reagents and materials, the apparatus, the drawing of the standard curve, sample preparation, the measuring procedure, the calculation of results, the repeatability requirement and the test report.

Laboratories working on forestry biomass materials use it so that enzyme activity results can be compared. It was issued on 6 February 2018 and has applied since 1 September 2018.

Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.

1 Scope

This standard specifies the terms and definitions, principles, reagents and materials, apparatus and instruments used in the determination of cellulase activity under given conditions And equipment, standard curve drawing, sample preparation, measurement procedures, calculation of results, repeatability requirements, test reports.

This standard applies to the determination of cellulase activity in forestry biomass materials analysis.

2 Normative references

The following documents for the application of this document is essential. For dated references, only the dated version applies to this document. For undated references, the latest edition (including all amendments) applies to this document.

3 Terms and definitions

GB/T 30366-2013 defined and the following terms and definitions apply to this document. For ease of use, the following list is repeated GB/T 30366-2013 some of the terms and definitions.

3.1 Forestry biomass

Biomass produced during forestry production and processing. Mainly include forest products (such as wood, bamboo, rattan, etc.), forestry residues (such as branches Ah, sawdust, sawdust, tops, skin and truncated heads, harvesting residues and processing residues such as nutshell and kernels, papermaking waste, and discarded wood) Energy forest. [GB/T 30366-2013, Definition 2.1.3]

3.2 Cellulase activity unit

The phase was released from the sodium carboxymethyl cellulose (CMC-Na) solution at a concentration of 5 g/L per minute at 37 °C, pH 5.5 When the amount of reducing sugar in 1µmol glucose is one enzyme activity unit (U).

4 Principle

Cellulase at a certain temperature and pH conditions, the hydrolysis of carboxymethyl cellulose produced by cellobiose, glucose and other reducing sugars can be alkaline Under the conditions of 3,5-dinitrosalicylic acid (DNS) reduction, the generation of brown-red amino compounds, maximum absorption at 540nm wavelength, The amount of reducing sugar produced by enzymolysis within a certain range is proportional to the absorbance of the reaction solution, and the amount of reducing sugar produced can be measured by a colorimetric method Cellulase activity was calculated.

5 Reagents and materials

5.1 All reagents used in this standard unless otherwise specified, are of analytical grade, water are in line with the provisions of GB/T 6682 secondary water.
