Scope

GB/T 30744-2014 is the English-translated version of 深海微生物样品前处理技术规范.

GB/T 30744-2014 sets out how samples of deep-sea microorganisms are to be pre-treated.

Clause 1 states that the standard specifies the technical requirements, the working conditions, the on-site handling of deep-sea samples, the extraction of microbiological and genetic material, storage and data processing, and that it applies to the on-site handling, extraction and preservation of deep-sea microbial samples taken at depths of 1000 m or more.

The document follows the workflow in order, with clause 4 on reagents and materials, clause 5 on instruments and equipment, clause 6 on general provisions, clause 7 on sampling at the scene, clause 8 on the isolation of deep-sea microbial strains, clause 9 on the extraction of deep-sea microbial genome DNA, clause 10 on the extraction of deep-sea microbial metagenomic DNA and clause 11 on the preservation of deep-sea biological samples and microbial resources.

Four informative annexes follow: Annex A with the formats of nine record sheets, running from the deep-sea sampling record to the deposit records for deep-sea bacteria, actinomycetes and fungi and the metagenomic DNA library record, Annex B on pressure isolates, Annex C on the molecular identification of microbial strains, which includes a table of universal primers for the 16S rRNA gene, and Annex D on the construction of a metagenomic Cosmid library.

Clause 3 defines the deep sea as marine areas at a depth of 1000 m or more and sets out what counts as a microbial sample, taking in strains of bacteria, fungi and actinomycetes, DNA and RNA, and the deep-sea water, sediment and rock samples that contain such genetic material.

The same clause defines pre-treatment as the processes carried out on collected samples before deep-sea microbial research, namely on-site handling, strain isolation, genome and metagenome DNA extraction and the preservation of samples and microbial resources, and it defines the cold-adapted microorganism, which grows well below 37 degrees C with an optimum growth temperature at or below 25 degrees C, and the thermophilic microorganism, which grows well at 55 degrees C or above.

Clause 2 cites GB/T 12763.4 and GB/T 12763.6, two parts of the specification for oceanographic survey, and HY/T 058 on the archiving of survey observations.

It was proposed by the State Oceanic Administration, is administered by the National Standardization Technical Commission for Oceanography (SAC/TC283) and was drafted by the Third Institute of Oceanography, State Oceanic Administration. It was issued on 9 June 2014 and has applied since 1 October 2014.
