GB 19171-2003 specifies the quality requirements, test methods, inspection rules, and labelling, marking, packaging, transport and storage of the pure culture (spawn) of Agaricus bisporus (button mushroom), and applies to its production, sale and use.

It covers the three levels of propagation - stock culture in test tubes, pre-culture spawn and cultivation spawn - with sensory requirements for each (container, plug, fill, inoculum, mycelium appearance, exudates, contaminants, antagonism, odour), microbiological requirements and growth rates at 24 °C ± 1 °C: 15 to 20 days to cover the slant for stock culture, and not more than 45 days to fill the container for pre-culture spawn and spawn.

Stock cultures must also have their esterase (Est) isoenzyme type identified by slab polyacrylamide gel electrophoresis (types G, H, HG1 to HG2 and HG4, with Rm values given for each band) and pass a fruiting test: germination within 24 h, pinning 12 to 16 days after casing, harvest at 18 to 22 days, flushes 7 to 10 days apart and biological efficiency not below 3 %.

Test methods cover sensory, microscopic and cultural inspection, growth rate, the electrophoresis procedure and a comparative cultivation test against the parent strain; sampling rates are 10 %, 5 % and 1 % by level.
