Scope
YY/T 1465.7-2021 is the English-translated version of 医疗器械免疫原性评价方法 第7部分:流式液相多重蛋白定量技术.
YY/T 1465.7-2021 is the Chinese pharmaceutical industry standard that gives the method for measuring several soluble proteins at once in a sample by multiplex flow cytometry, using bead arrays.
It is the seventh part of the YY/T 1465 series on the immunogenic evaluation of medical devices and is used to judge the immune response a device or material provokes: the standard sets out the samples and their handling, the bead and antibody reagents, the instrument settings and calibration, the running of the assay and the calculation of the concentrations from the standard curve. Issued on 9 March 2021, it has applied since 1 April 2022.

Foreword
YY/T 1465 "Methods for Evaluation of Immunogenicity of Medical Devices" is divided into the following parts.
---Part 1: In vitro T lymphocyte transformation test;
---Part 2: Determination of serum immunoglobulin and complement components (ELISA method);
---Part 3: Plaque forming cell determination agar solid phase method;
---Part 4: Mouse peritoneal macrophages phagocytize chicken red blood cells in vivo method;
---Part 5: Use M86 antibody to determine the clearance rate of alpha-Gal antigen in animal-derived medical devices;
---Part 6: Determination of animal spleen lymphocyte subsets by flow cytometry;
---Part 7: Flow cytometry multiple protein quantification technology. This part is part 7 of YY/T 1465.
This section was drafted in accordance with the rules given in GB/T 1.1-2009.
Introduction
Immune response is an important defense mechanism of the body. As an exogenous substance, medical devices pass through various ways after being in contact with the human body.
It affects the immune response of the body's immune system, especially for animal-derived medical products, allogeneic products, and tissue-engineered medical products Wait. Although the interaction between medical devices/materials and the immune system may produce different immune responses, they can be roughly divided into two types.
That is, humoral immune response and cell-mediated immune response. At present, it is still impossible to determine whether the immune response generated by medical devices or materials stimulates the host.
It is beneficial or harmful. Therefore, it is very important to use medical devices/materials to conduct immune response research to obtain relevant information.
GB/T 16886.20 gives guidelines for possible immune reactions and potential immune toxic reactions that may occur when medical devices come into contact with the human body.
But there is no specific test method. YY/T 1465 series standards are expected to provide specific test methods for the implementation of GB/T 16886.20.
This part of YY/T 1465 provides the simultaneous quantitative detection of multiple soluble proteins in samples using the flow-type liquid phase multiple protein quantification technology The method provides specific test methods for evaluating the potential of medical devices/materials to stimulate the body's immune response, which can be used as GB/T 16886.20 An alternative method standard in immunotoxicology tests. Other methods confirmed to be applicable can also be used.
1 Scope
This part of YY/T 1465 stipulates the quantitative detection of a variety of soluble proteins in samples using flow cytometry multiple protein quantification technology. experiment method.
This section is suitable for evaluating the immune response induced by medical devices/materials.

3 Flow-type liquid phase multiple protein quantification technology
3.1 Experimental principle
The flow cytometry liquid phase multiple protein quantification technology is based on the combination of fluorescent microsphere technology and "double antibody sandwich" liquid phase detection technology, which can be realized Simultaneous quantitative detection of multiple soluble proteins. The microspheres are respectively labeled with different wavelengths of fluorescence, and each type of microsphere is respectively coupled to the corresponding target antigen The capture antibody specifically captures the target protein in the sample or standard, and the capture microspheres, target protein and the added detection antibody form a double The "sandwich" structure complex of antibody sandwich, the fluorescence intensity of the complex is detected by flow cytometry. At the same time, through traceability The standard curve is drawn with the standard product, which can realize the quantitative detection of multiple soluble proteins in the sample to be tested at the same time.
3.2 Experimental animals
3.2.1 General
All animal experiments should be carried out in laboratories approved by the national accreditation body and in compliance with all applicable regulations on the welfare of laboratory animals. It should meet the requirements of GB/T 16886.2.
3.2.2 Species and requirements of animals
Commonly used experimental animals are mice. In this section, it is recommended to use healthy BALB/c mice that have not been tested and are free of specific pathogens.
(SPF) grade, 6 to 8 weeks old. The animals should be kept for at least 5 days before the formal test to adapt to the laboratory environment. Laboratory animals should be marked with species and product Department, source, gender, weight and age in weeks. At the beginning of the experiment, the animal's weight difference should be controlled to the minimum range, and the weight of each animal should not exceed Over ±20% of the average weight range of animals of the same sex. It is recommended to use at least 3 mice of each sex.
If other species of animals are selected, their suitability should be explained.
3.3 Sample preparation
The test sample preparation is carried out in accordance with the principles of GB/T 16886.12.
