Scope
GB/T 40049-2021 is the English-translated version of 鸡肠炎沙门氏菌PCR检测方法.
GB/T 40049-2021 is the Chinese national PCR method for detecting Salmonella Enteritidis carried by chickens of all ages and by chicken products, and for its molecular typing.
It defines the term housekeeping gene and the abbreviations DNA and PCR used in the method.
It lists the reagents and materials, among them LB liquid medium, buffered peptone water for pre-enrichment, selenite cystine and tetrathionate brilliant green enrichment broths, XLD agar, TAE running buffer and the positive and negative control strains, with their preparation given in an annex.

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
1 Scope
This standard specifies the PCR detection method for molecular typing of Salmonella enteritidis.
This standard is applicable to the PCR method detection of Salmonella enteritidis carried in chickens and their products of various ages.

5 Reagents and materials
Unless otherwise specified, all reagents used are of analytical grade. The experimental water meets the GB/T 6682-2008 Grade II water regulations.
5.1 LB liquid medium (see A.1 in Appendix A for the preparation method).
5.2 Buffered peptone water (BPW) pre-enrichment solution (see A.2 for the preparation method).
5.3 Selenite cystine (SC) enrichment solution (see A.3 for the preparation method).
5.4 Tetrathiosulfonate brilliant green (TTB) bacteria enrichment solution (see A.4 for the preparation method).
5.5 Xylose Lysine Deoxycholate (XLD) Agar D.3 (see A.5 for the preparation method).
