Scope

GB/T 22101.5-2009 is the English-translated version of 棉花抗病虫性评价技术规范 第5部分:黄萎病.

This part of GB/T 22101 sets the method for identifying the resistance of cotton to verticillium wilt and the criteria for evaluating it. The scope clause is mangled in the draft, but its second sentence states that the part applies to the identification and the rating of the resistance of cotton to verticillium wilt.

The foreword places it as part 5 of GB/T 22101, whose other parts cover cotton bollworm, aphids, pink bollworm and wilt, and records that Appendix B is normative and Appendix A informative.

Clause 2 carries two definitions whose term names the draft does not print: one a percentage taken on the total number of cotton seedlings, the other a composite index built from morbidity and severity.

Clause 3 covers the culture of the pathogen, listing the basic equipment, which includes an incubator, a clean bench, an autoclave, a refrigerator, a temperature-controlled greenhouse held between 20 and 28 degrees Celsius, an aluminum pan, an electric oven, dishes, tubes, scissors, tweezers, jars and alcohol lamps, and describing a culture on sand grains, cottonseed or wheat grains that are boiled, drained, loaded into jars, autoclaved for 2 h, inoculated on the clean bench from an existing flat or ramp culture, and then held in an incubator at 25 degrees Celsius for 10 to 15 days.

Clause 4 covers the establishment of the artificial verticillium wilt nursery, which is to be set up in an area where the disease occurs uniformly, where the disease index of the susceptible control in a normal year falls between 40.0 and 60.0, where climatic conditions have little effect and virulent strains are widely distributed, the standard pointing to areas north of 38 degrees latitude where the July and August average temperature exceeds 28 degrees Celsius on fewer than 20 days; the identification itself is described as adult plant identification in an artificial disease nursery field.

The same clause has the strain chosen from the locally dominant flora, 450 kg to 750 kg of well grown culture spread evenly over each hectare and plowed in two or three times so that it mixes with the soil, and the nursery brought to a disease index of about 50 on the susceptible variety in each plot, after which the cotton stalks are crushed and returned to the field according to the incidence of the year.

The page shows no earlier edition; it was issued on October 30, 2009 and has applied since December 1, 2009.

Foreword

GB/T 22101 "cotton pest resistance evaluation of technical specifications" is divided into five parts.

--- Part 1. cotton bollworm;

--- Part 2. aphids;

--- Part 3. pink bollworm;

--- Part 4. wilt;

--- Part 5. Verticillium wilt.

This part of GB/T 22101 Part 5.

Appendix B to this part of the normative Appendix Appendix A is an information appendix.

This part of the People's Republic of China Ministry of Agriculture and focal points.

1 Scope

Methods and evaluation criteria set resistance.

This section applies to cotton resistance to Verticillium wilt resistance identification and assessment.

2 Terms and definitions

The following terms and definitions apply to this part of the GB/T 22101 of.

2.1 Cotton seedlings incidence percentage of total number of cotton seedlings

2.2

A comprehensive assessment of composite indicators of morbidity and severity.

3 Training pathogens

3.1 Training pathogens basic equipment

Incubator, clean table, autoclaves, refrigerators, temperature-controlled greenhouse (keeping the temperature between 20 °C ~ 28 °C), aluminum pan, electric Oven, dish, tube, scissors, tweezers, jars, lamps and other alcohol.

3.2 Cotton verticillium wilt pathogen culture

Verticillium fungus (see Appendix A) bacteria cultures using sand grains or cottonseed, cottonseed or wheat grains boiled first rose up, drain the water After hours, loaded jars, autoclaved 2h; clean table in good verticillium wilt bacteria had been cultured flat or ramp access which, with the After placed in 25 °C incubator cultured 10d ~ 15d.

4 Verticillium establishment of nurseries

4.1 Nurseries requirements

Artificial verticillium wilt nursery should be established in a suitable area that is conducive to regional verticillium wilt occur uniformly incidence; requires a normal year, susceptible control Condition index of between 40.0 to 60.0, less affected by climatic conditions; should virulent strains of widely distributed appropriate.

Identification of resistance Verticillium using "artificial disease nursery field of adult identification of" identification. Suitable areas for the summer in July and August level Average temperatures of over 28 °C for less than 20d, 38 degrees north latitude regions.

4.2 Identification of Pathogens used species

According to the local advantage throughout the flora, the appropriate choice of virulent strains.

4.3 The establishment of nurseries

Per hectare inoculation 450kg ~ 750kg will develop good bacteria uniformly applied into the field, then plowed over 2 to 3 times, so that bacteria And soil mix evenly. In susceptible varieties in nurseries each cell uniform onset disease index of about 50 to. After the establishment of nurseries, according to when The incidence of disease in the current year will be crushed tieback cotton stalks.
