Scope

GB/T 21679-2023 is the English-translated version of 法庭科学 DNA数据库建设规范.

GB/T 21679-2023 is the Chinese criterion for building and running a forensic science DNA database.

It sets the overall requirements and then the requirements for the networked laboratories, the users, the detection reagents and equipment, the collection and storage of information, the comparison of data, the use made of comparison results, quality control and security.

Its terms clause defines locus, short tandem repeat, mitochondrial DNA, hypervariable region, DNA database, the database management organization, the technical person in charge and the database administrator.

It applies to the construction of forensic science DNA databases and to the design, development and testing of the software behind them, and it cites GB/T 22239, GB/T 28448, GB/T 41009, GB/T 41615 and four GA/T laboratory standards.

It replaces GB/T 21679-2008, was issued on 28 December 2023 and has applied since 1 July 2024.

Foreword

This document complies with the provisions of GB/T 1.1-2020 "Standardization Work Guidelines Part 1: Structure and Drafting Rules of Standardization Documents" Drafting.

This document replaces GB/T 21679-2008 "Specifications for the Construction of Forensic Science DNA Database" and is consistent with GB/T 21679-2008.

In addition to structural adjustments and editorial changes, the main technical changes are as follows:

---Changed the terms and definitions of "DNA database" and "DNA database administrator" (see 3.5, 3.10, 2.1, 2008 version) 2.9);

---Added the terms "locus", "short tandem repeat sequence", "mitochondrial DNA", "hypervariable region" and "DNA database management organization" "DNA database network laboratory" "Technical person in charge" "DNA database user" "DNA detection reagent" "DNA detection equipment" "Prepare" (see 3.1~3.4, 3.6~3.9, 3.11, 3.12);

---The terms "basic DNA database", "personnel sample library", "on-site evidence library", "kinship sample library" and "large disaster accident sample library" have been deleted. "This database", "Classification of DNA database construction objects" and "Relationship attributes of DNA database construction samples" (see the 2008 edition of 2.2~2.8);

---Added abbreviations (see Chapter 4);

---Deleted "Function, Structure and Responsibilities" (see Chapter 3 of the 2008 edition);

---Deleted "sample collection for database construction objects" (see Chapter 4 of the 2008 edition);

---Added "overall requirements" (see Chapter 5);

---Added the content of "Networked Laboratory" (see Chapter 6);

---Added "user" content (see Chapter 7);

---Changed the contents of reagents, equipment, loci, information collection and data comparison involved in the library construction process (see Chapter 8 ~ Chapter 11, Chapter 5~Chapter 8 of the 2008 edition);

---Added the content of "application of comparison results" (see Chapter 12);

---Added the content of "Quality Control" (see Chapter 13);

---Added "security" requirements (see Chapter 14).

1 Scope

3 Terms and definitions

The following terms and definitions apply to this document.

3.1 Locus

The location occupied by a gene on a chromosome or a segment of genomic DNA. [Source: GA/T 1972-2021,3.5]

3.2 Short tandem repeats

Repeating units are composed of 2 to 6 nucleotides, and DNA fragments are repeated 5 to 60 times, with a total length of less than 400 bp. DNA sequence.

Note: In the human genome, according to the type of chromosome, it is divided into autosomal STR, Y chromosome STR and X chromosome STR. [Source: GA/T 1972-2021, 3.10, modified]

3.3 Mitochondrial DNA

Closed circular double-stranded DNA found in the mitochondria of human cells.

Note: Human mitochondrial DNA is about 16.5Kb; it does not follow the Mendelian laws of inheritance and is maternally inherited. The genetic information is transmitted to the egg through the egg cell.

Offspring; no cycle changes of mitosis and meiosis; genetic material is located in organelles and is not affected by nuclear transfer; mtDNA copies in single cells
