6.3 Stool or anal swab sample. collect a stool sample, add an equal volume of sterilized physiological saline to grind the homogenate, and centrifuge at 4 °C 5 000 r/min 15 minutes, collect the supernatant for testing. Collect anal swab samples, infiltrate it with a small amount of sterile physiological saline, and take the supernatant for testing.
6.4 Tissue samples. Collect tissue samples such as intestines and stomach, add an equal volume of sterilized physiological saline to grind the homogenate, and centrifuge at 4 °C 5 000 r/min 15 minutes, collect the supernatant for testing.
7 ELISA detection method
7.1 Operation steps
7.1.1 Coating antigen. Dilute the purified VP6 protein with the coating solution to 3 µg/mL, 100 µL per well, and coat the microtiter plate. After sealing, Coating at 2 °C~8 °C for 12 h. The ELISA detection method can use an equivalent commercial ELISA antibody detection kit, according to the kit instructions Book to operate.
7.1.2 Discard the liquid in the wells, add about 300 µL of washing solution to each reaction well for washing, wash 5 times, the last time in a clean suction Pat dry the plate on water paper.
7.1.3 Blocking. add 250 µL of blocking solution to each well and place it in a 37 °C incubator for 2 h.
7.1.4 Repeat step 8.1.2 and store at 4°C for later use.
7.1.5 Dilute the serum to be tested 1.100 with the sample diluent, add 100 µL to each well, and add the negative control and positive control serum at the same time.
2 wells, 100 µL per well. Place in a 37 °C incubator to react for 1 h.
7.1.6 Repeat step 8.1.2.
7.1.7 Dilute the anti-pig PRV enzyme-labeled antibody to the working concentration, add 100 µL to each well, and place in a 37 °C incubator to react for 1 h.
7.1.8 Repeat step 8.1.2.
7.1.9 Color development. add 100 µL of substrate color development solution to each well, shake gently, and develop color at 37 °C in the dark for 10 min.
7.1.10 Stop the reaction. add 100 µL stop solution to each well to stop the reaction.
7.1.11 Reading. Read the absorbance value at 450 nm on the microplate reader.
7.2 Judgment of results
7.2.1 Detect the light absorption value (OD450) of each well on the microplate reader, calculate the average OD450 value of the positive control and the average value of the negative control.
Average OD450 value. The average OD450 value of the positive control serum is greater than 0.6, and the average OD450 value of the negative control serum is less than 0.16.
Established.
7.2.2 When the ratio of the average OD450 value (P) of the sample well to the average OD450 value (N) of the negative control is greater than or equal to 2.0 (ie P/N >= 2.0), it was judged to be PRV antibody positive.
7.2.3 When the average OD450 value (P) of the sample well and the average OD450 value of the negative control...
