GB/T 45033-2024 gives the Chinese method for determining Candida utilis in microbial feed additives. Live yeast preparations are sold on a declared viable count, and the declaration is the product: the buyer cannot see it, the count falls during storage and transport, and a method that does not distinguish the declared organism from the other yeasts and bacteria present cannot support the claim. The standard sets the reagents and materials, the instruments and equipment, the sampling, and then the test procedure: the preparation and dilution of the sample, the selective media and the culture conditions under which Candida utilis grows and competitors are suppressed, the incubation, the counting of the colonies and their confirmation by morphological and biochemical characteristics or by molecular identification where required, and then the calculation, the expression of the result, the precision and the test report. It takes effect on 1 June 2025.

This document describes a method for the determination of the count of Candida utilis in microbial preparations for feed. This document applies to the determination of feed microbial preparations containing Candida utilis.

4 Reagents or materials

Unless otherwise specified, only analytical grade reagents were used.

4.1 Water. GB/T 6682, grade 3. 4.2 0.85% sterile saline. Accurately weigh

8.5 g of sodium chloride and add it to 1000 mL of water. Stir until completely dissolved. After packaging, place the mixture at 121 °C. Sterilize for 30 min and set aside.

4.3 Red Bengal agar medium. see Appendix A, A.1.

4.4 Yeast peptone dextrose (YPD) agar medium. see A.2.

5.Instruments and equipment In addition to routine sterilization and culture equipment in microbiology laboratories, other instruments and equipment are as follows:

5.1 Incubator. controllable accuracy ±1 °C.

5.2 Balance. sensitivity is

0.1 g.

5.3 Optical microscope. 400x to 1000x.

5.4 Slap-type homogenizer and sterile homogenizing bag.

5.5 Vortex mixer.

5.6 Sterile culture dish. 90 mm in diameter.

5.7 Polymerase chain reaction (PCR) instrument.
