1 Scope

This document specifies the principles, reagents and methods for the determination of lycopene, lutein and carotene in tomato products by ultra-high performance liquid chromatography.

Materials, equipment, analytical procedures, calculation of results, precision, limits of detection, and limits of quantification.

This document applies to lycopene, lutein, alpha-carotene and beta-carotene in tomato powder, tomato paste, tomato beverages, and dried tomato products element determination.

4 Principles

After adding water and ethanol to the sample, extract the lycopene, lutein and carotene in it with ether-n-hexane-cyclohexane mixed solution.

Separation by high performance liquid chromatography, detection by UV detector or diode array detector, and quantification by external standard method.

Unless otherwise specified, use only chromatographically pure reagents.
