Scope

GB/T 35098-2018 is the English-translated version of 微束分析 透射电子显微术 植物病毒形态学的透射电子显微镜鉴定方法.

GB/T 35098-2018 is the Chinese national method for identifying plant viruses from their morphology under the transmission electron microscope.

It sets out the principles, instruments, reagents and materials, environmental conditions, sample preparation and identification steps, the quality requirements for the identification and how the identification report is issued, together with decontamination treatment.

It defines terms such as virus, virion, capsid, nucleocapsid, inclusion body and antigen. Informative annexes give formulations of commonly used reagents and the morphological and cytopathological features of the families and genera of plant viruses.

It applies to observation with all types of transmission electron microscope and is used by laboratories that diagnose plant virus diseases. It was issued on 14 May 2018 and has applied since 1 April 2019.

Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.

Introduction

Plant viruses are an important class of pathogens that infect angiosperms, gymnosperms and ferns, causing crops worldwide.

Diseases of fruits, fruit trees, flowers, pastures, and medicinal plants cause a decline in yield and quality, which seriously affects human production and life..2016 International The plant virus announced by the International Committee for Classification (Taxonomy of Viruses, ICTV) involves 4 Head, 26 families, 5 subfamilies, 118 genera, a total of 1323 species. Virology morphology is an important basis for virus identification and classification, based on viral particles The shape, size, presence or absence of the capsule, the symmetry of the capsid, the cytopathology of the virus in the host, and the genomic sequence Equivalent molecular biology evidence can be used to diagnose and identify viruses to families, genera, and species. Since the individual of the plant virus is as small as several tens of nanometers, it can be transmitted by electrons.

The microscope can observe its morphology. In order to standardize the technical methods of plant virus morphology detection, correctly guide the diagnosis of plant virus pathogens And identification work, it is necessary to develop a national standard for morphological identification methods of plant virus transmission electron microscopy.

1 Scope

This standard specifies the steps, quality requirements and identification reports for morphological observation and identification of plant viruses using transmission electron microscopy. The announcement of the announcement.

This standard applies to the observation and identification of plant virus morphology by various types of transmission electron microscopy.

3 Terms and definitions

The following terms and definitions apply to this document.

3.1 Virus

A pathogen consisting of a nucleic acid and a protein coat and having an invasive activity and capable of self-replication in a cell.

3.2 Virion

A mature or structurally intact, infectious virus individual.

3.3 Capsid

A symmetrical protein envelope consisting of a collection of protein subunits and surrounding the nucleic acid genome.

3.4 Nucleocapsid

A combination of a protein capsid surrounding a viral nucleic acid and a nucleic acid.

3.5 Inclusion body

An abnormal structure composed of viruses or cellular components produced in a host cell after infection by a virus.

Note: The inclusion bodies show different morphological features under optical or TEM.

3.6 Antigen

A substance that stimulates the body of an animal to produce antibodies and sensitize lymphocytes and to react specifically with it.
