YC/T 31 Preparation of Tobacco and Tobacco Product Samples and Determination of Moisture Content by Oven Drying Method

3 Terms and Definitions

This document does not contain any terms or definitions that need to be defined.

4 Principles

The sample was sequentially treated with 80% ethanol-saturated sodium chloride solution to remove sugar and 90% dimethyl sulfoxide-water solution to remove starch and other interferences before being added...

The polymer was depolymerized in 72% sulfuric acid solution, diluted with water, and then subjected to high-temperature acid hydrolysis. The mixture was then separated by filtration to obtain the acid hydrolysis filtrate. The acid hydrolysis filtrate was analyzed by ion chromatography.

The composition and concentration of monosaccharides in the acid hydrolysis products are used to calculate the cellulose content, and the concentrations of arabinose and galactose in the acid hydrolysis products are used to determine the cellulose content.

The concentrations of sugar, xylose, and mannose are used to calculate the hemicellulose content.

5 Reagents

5.1 Water, with a resistivity of not less than 18.0 Mohm·cm.

5.2 L-()-arabinose, D-galactose, D-()-xylose, D-mannose, and glucose, all with a purity of not less than 99%.

5.3 Anhydrous ethanol.

5.4 Dimethyl sulfoxide.

5.5 Concentrated sulfuric acid, 98% (mass fraction).

5.6 Sodium hydroxide solution, 50% (mass fraction).

5.7 Sodium acetate, purity >=99%.

5.8 Sodium chloride.

5.9 80% ethanol-saturated sodium chloride solution. Weigh 64 g of sodium chloride (5.8) into a 1000 mL beaker, add.200 mL of water to dissolve, then...

Add 800 mL of anhydrous ethanol (5.3), mix well, cover and let stand for later use.

5.10 90% Dimethyl Sulfoxide-Aqueous Solution. Transfer 900 mL of dimethyl sulfoxide (5.4) into a 1000 mL volumetric flask and dilute to volume with water.
