3 Method summary

The sample was diluted, centrifuged at high speed, and determined by liquid chromatography-mass spectrometry/mass spectrometry and quantified by external standard method.

4 Reagents and materials

Unless otherwise stated, the reagents used are of excellent grade and the water is the first grade water specified in GB/T 6682.

4.1 Acetonitrile. chromatographically pure.

4.2 Formic acid.

4.3 5% acetonitrile-0.1% formic acid solution. Accurately measure 50mL acetonitrile, 1mL formic acid, dilute to 1L with water, mix and use.

4.4 0.1% formic acid-water solution. Accurately measure 1 mL of formic acid, dilute to 1 L with water, mix well and set aside.

4.5 Natamycin standard (CAS number 7681-93-8, molecular weight. 665.73). purity >= 89.9%.

4.6 Natamycin standard stock solution. accurately weigh the appropriate amount of natamycin standard, and use methanol to prepare a standard of 1.0mg/mL.

Stock solution.

4.7 Natamycin standard intermediate solution. Accurately absorb the appropriate amount of natamycin standard stock solution (4.6) with 5% acetonitrile-0.1% formic acid solution (4.3) A standard intermediate solution having a concentration of 10 µg/mL was prepared.

4.8 Natamycin standard working solution. Accurately absorb the appropriate amount of natamycin standard intermediate solution (4.7), using 5% acetonitrile-0.1% formic acid solution (4.3) The natamycin standard intermediate solution was diluted stepwise to prepare a standard working solution of 0.2 µg/L to 100 µg/L.

5 Instruments and equipment

5.1 High Performance Liquid Chromatography-Mass Spectrometry/Mass Spectrometer. Triple Quadrupole Mass Spectrometry with Electrospray Ion Source (ESI).

5.2 Analytical balance. Sensing amount 0.01mg.

5.3 Vortex mixer.
