3 Method summary
After the sample is diluted with buffer solution, the sample is adjusted to neutrality with sodium hydroxide solution, purified by an immunoaffinity column, and finally colored by liquid phase.
Spectrum-tandem mass spectrometer detection, external standard method quantification.
4 Reagents and materials
Unless otherwise stated, all reagents were of analytical grade and water was a grade of water as specified in GB/T 6682.
4.1 Methanol. Chromatographically pure.
4.2 Acetonitrile. Chromatographic purity.
4.3 acetic acid. chromatographic purity.
4.4 Sodium bicarbonate.
4.5 Sodium chloride.
4.6 Sodium hydroxide.
4.7 Ammonium acetate.
4.8 Acetonitrile solution (50%). Pipette 100 mL of acetonitrile (4.2) and dilute to.200 mL with water.
4.9 Acetic acid solution (0.1%). Pipette 1.0 mL of acetic acid (4.3) and dilute to 1 L with water.
4.10 Buffer solution. Accurately weigh 10g of sodium bicarbonate (4.4) and 75g of sodium chloride (4.5), dissolve in water and bring to volume to 500mL.
4.11 sodium hydroxide solution (2%). accurately weighed 20g of sodium hydroxide (4.6), dissolved in water and set to 1L.
4.12 Ammonium acetate solution (0.2mol/L). Weigh 15.4g of ammonium acetate solid (4.7), dissolve with water and make up to 1L.
