1 Scope

This standard specifies the juice of cyclic fatty acid bacillus (Alicyclobacillusspp.) Test method.

This section applies to fruit and vegetable juices cyclic fatty acid bacillus test.

3 Terms and Definitions

The following terms and definitions apply to this document.

Alicyclobacillus Bacillus Alicyclobacillusspp.

Alicyclobacillus is a group of heat-resistant Bacillus, Bacillus acidophilus rod-shaped, Gram-positive to the variable, its pH range 2.0 to 6.0 and a temperature of 45 °C when good growth.

4 Equipment and Materials

In addition to the microbiological laboratory conventional sterilization and cultivation equipment, other equipment and materials as follows.

4.1 Balance. a sense of the amount of 0.1g.

4.2 Refrigerator. 2 °C ~ 5 °C.

4.3 incubator. 45 °C ± 1 °C, 65 °C ± 1 °C.

4.4 constant temperature water bath. 80 °C ± 1 °C, 45 °C ± 1 °C.

4.5 phase contrast microscope. 10 × ~ 100 ×.

4.6 sterile conical flask. 500mL, 250mL.

4.7 sterile petri dish. diameter of 90mm.

4.8 sterile pipette. 1mL (with 0.01mL scale), 10.0mL (with 0.1mL scale) or micro pipettes and tips.

4.9 pH meter or pH colorimetric tubes or precision pH test paper.

4.10 Membrane. Diameter 0.45µm.

4.11 a vortex mixer.
