1 Scope

This standard specifies the phytosanitary Apple stem grooving virus (Applestemgroovingvirus) of quarantine and identification methods.

This standard applies to seedlings of apples, pears, etc., quarantine and identification of other propagating material apple stem grooving virus.

3 Reagents and solutions

Unless otherwise specified, all test reagents were analytical grade.

3.1 ELISA reagents. See Appendix B, Section B. Chapter 1.

3.2 The total RNA extraction reagent. TrizolReagent.

3.3 RT-PCR step reaction reagents. Ready-to-GoRT-PCRBeads.

3.4 chloroform.

3.5 isopropanol.

3.6 75% ethanol.

3.7 ethanol.

3.8 RNase-free water. double distilled water or commodities by 0.1% DEPC (diethylpyrocarbonate) treated RNase-free water.

3.9 reverse transcriptase and buffers.

3.10 ribonuclease inhibitor.

3.11 dNTPs. containing dATP, dCTP, dGTP, dTTP.
