3 Material preparation
3.1 Instruments and equipment
3mm thick 100mm × 45mm glass plate; voltage 0V ~ 300V, current 0mA ~ 50mA electrophoresis instrument; width 100mm Electrophoresis tank; 3mm diameter puncher; 100µL micro-adjustable pipette and matching tip; plastic capillary blood collection tube.
Barbital sodium 10.31g Tris-hydroxymethylaminomethane 6.005g Sodium azide 0.2g Secondary water (in accordance with GB/T 6682) 1000mL After adding the appropriate amount of secondary water (according to GB/T 6682), dissolve the pH value to 8.5-8.6 with hydrochloric acid, and finally use the second grade.
Water (in accordance with GB/T 6682) to a volume of 1000mL.
3.3 Agarose
Analytically pure agarose for domestic or imported electrophoresis.
3.4 Antigen
It was prepared from a cat kidney passage cell culture of the Aleutian virus international standard strain Utah-1. Provided by the designated unit.
3.5 Serum
3.5.1 Positive serum. provided by the designated unit.
3.5.2 Negative serum. provided by the designated unit.
3.5.3 Serum to be tested. Blood is collected from the tip of the toe with a plastic capillary tube. The two ends of the tube are sealed and sealed, placed in an environment of 20 ° C to 30 ° C for 1 h, and then placed.
4 ° C refrigerator 18h ~ 20h, precipitated serum, add 0.5mL or 1.5mL centrifuge tube, centrifuge at 3000r/min for 5min, take the supernatant spare.
4 Operating procedures
4.1 Boarding
Take 3mm thick 100mm × 45mm horizontal scratch-free glass plate, wash the oil, put it on the horizontal table, use 10mL the pipette draws 0.8% agarose buffer which has been dissolved and cooled to about 70 ° C, and is evenly spread on a glass plate with a thickness of 3 mm.
When the glass plate is surrounded by a baffle, it will be used after cooling and solidification, and it will be used up on the same day.
