Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009 "Guidelines for Standardization Work_Part 1. Structure and Preparation of Standards" this standard replaces JY 83-82 "Technical Conditions for Plant Cell Mitosis (Slice of Onion Root Tips)". In addition to editorial changes, the main the technical changes are as follows.

-Add product model name;

-Eliminate the "slice thickness within 5 microns" which is difficult to inspect;

-Elimination of single dyeing requirements;

-Cancel the classification of the original qualified products into first-class products and second-class products.

1 Scope

This standard specifies the type designation, requirements, test methods, inspection rules, marks, instructions, Packaging, transportation and storage.

This standard applies to mitotic slide specimens of plant cells for teaching in primary and secondary schools.

3 Model designation

4 Requirements

4.1 Slides and coverslips, appearance, specimen collection and processing, specimen staining, and specimen covers should comply with JY/T 0067-2011 Chapter 4.

4.2 Specimens should be taken from the root tip of onions during the period of vigorous cell division, and the upper end of the root should be neat.

4.3 Make a longitudinal cut, and place 1 to 3 pieces of material on each slide.

4.4 Cells in the pre-, middle, late, and terminal stages of division should be clearly displayed.

4.5 The morphological characteristics of chromosomes at each stage of division are typical.

4.6 The root apex should be intact and not damaged, and there should be no obvious cracks between the cells and obvious contraction of the nucleus.

4.7 The nucleus, nucleoli and chromosome should be clearly colored, and the cytoplasm should be pale.

5 Test method

5.1 According to the provisions of Chapter 5 of JY/T 0067-2011.

5.2 Sections 4.3 and 4.5 of this standard are tested at a microscope magnification of 40 times, and sections 4.4 and 4.7 at a microscope magnification of 400 Inspection at times.

6.1 Implementation of the relevant provisions of Chapter 6 of JY/T 0067-2011.

6.2 In this standard, 4.3, 4.4, 4.5, 4.6 and 4.7 are the main performance indicators.
