Structure and Preparation of Standards" this standard replaces JY 67-1982 "General Technical Conditions for Biological Slide Specimens". In addition to editorial changes, the main technical changes are as follows.

-Add definition of main types of slide specimens;

-Increase environmental requirements and improve packaging requirements;

-Cancel the classification of the original qualified products into first-class products and second-class products.

3 Terminology

This standard uses the following terms.

Prepared biological microscope slides Individual organisms, organs, tissues or cells are appropriately processed and encapsulated in microscope slides (hereinafter referred to as slides) with a capping agent and Permanently preserved specimens formed between microscope coverslips (hereinafter referred to as coverslips).

Sections

A slide specimen prepared by cutting a biological tissue into thin slices. Cross section, that is, the cutting direction is perpendicular to the long axis of the material or structure or fiber direction; vertical section, that is, the cutting direction is parallel to the long axis of the material or structure, or fiber direction;

With or without specific requirements.

Whole mounts

Tiny biological entities or structures that do not need to be cut into thin slices and directly encapsulated slide specimens after proper processing. Squeezing or squeezing material the finished packing is called pressing; the packing made by separating materials is called separating loading.

Smear smear

Liquid materials or sticky materials are smeared on glass slides when fresh, and slide specimens made after proper processing.

Grinding Ground

For hard materials such as teeth and bones, grind on the abrasive tool into a nearly transparent sheet, and then make a glass slide specimen after proper processing.

4 Requirements

4.1 Use environmental conditions

Temperature. -10 °C ~ 40 °C, relative humidity. not more than 90%.

4.2 Slides and coverslips

4.2.1 Dimensions of slides and coverslips

4.2.1.1 The overall dimensions of the slide shall comply with the provisions in Table 1 of JB/T 8230.3-1997, Section 4.1. The basic dimensions shall be 76mm in length and 26mm in width.

The dimensions of the glass slide for a specific product shall be specified separately in the product standard.

4.2.1.2 Except for the outer dimensions of the coverslip, which should comply with the provisions in Table 1 of JB/T 8230.4-1997 Section 4.1, square coverslips should be used.

For side lengths of 16mm and 14mm, round coverslips should be 20mm and 14mm in diameter. The thickness of the coverslip should conform to JB/T Type 8230.4-1997, Table 2, Table 2, Type G.

4.2.2 Appearance of slides and coverslips

4.3.1 The typical biological material and the correct material extraction site should be selected according to the tissue structure to be displayed by the product.

4.3.2 The material should be fresh and timely, and the tissue should not have autolysis, lesions and shrinkage.

4.3.3 General plant tissues should not be too young to keep clear cell boundaries, normal morphology, and clear staining. Have specific observations What is required is not limited.

4.3.4 If there is no specific requirement, the thickness of the section is generally based on the non-overlapping phenomenon of cells. Consecutive section specimens should be of consistent thickness Continued, there should be no shortage.

4.3.5 The sections should be free of obvious knife marks, cracks, damages, wrinkles, and the sections should be flat.

4.3.6 Loaded specimens should be complete in shape, normal in shape, obvious in characteristics, suitable in pigments, no air in the body, and trimmed neatly and smoothly at the edges.

Special requirements shall be specified separately in the product standard.

4.3.7 Smears, tablets, and separated specimens should be evenly spread and the cells dispersed, and the number should not be scarce.

4.3.8 The thickness of tooth, bone and other abrasive specimens should be moderate to make the relevant tissue structure clear.

4.4 Staining of specimens

4.4.1 Except that the specimen itself can meet the observation requirements, the specimen needs to be stained, and the tissue structure to be displayed should be colored, clear and uniform.

It should be uniform and coordinated, and there should be no deep inside or shallow inside.

4.4.2 There should be no deposits or stains on the specimen.

4.4.3 In addition to achieving a specific effect, a permanent and fade-free stain and dyeing method should generally be selected.

4.5 Covering of Specimens

4.5.1 Specimens should be sealed in the middle of the slide. Continuous sections and large-area specimens can be sealed on one side of the slide.

4.5.2 Coverslips of appropriate size should be selected for the cover, and the specimens should be covered at the moderate position of the coverslip. The coverslip should be larger than the cover.

Specimen. If a square cover glass is used, its four sides should be parallel to the edge of the slide.

4.5.3 When two specimens with vertical and horizontal sections are placed on the same slide, the size ratio should be appropriate and the thickness of the sections should be the same.

4.5.4 Specimens can be divided into back, ventral, front, and back. When above and below, the coverslips should be sealed with the back, front, and top facing.

Installed. When observing a specimen with a specific orientation, its requirements shall be separately stated in the product standard.

4.5.5 The cover glass is filled with a capping agent. The capping agent should not overflow, and there should be no air bubbles or foreign matter.

4.5.6 The capping agent after capping should be dry so that the specimen cannot be moved.

4.5.7 The coverslips of thicker specimens can be left with a uniform, dry cover agent around the cover glass, and its width should not be greater than 1.5mm.

4.5.8 The coverslips of specimens covered with water-soluble transparent agent (glycerin, etc.) should be sealed around the sealing ring.

For the thickness of the coverslip. When using double-layer coverslips, the two capping agents should not impregnate each other and there should be no turbidity.

4.5.9 The capping agent shall be made of natural resin or synthetic resin, which is colorless or light yellow, transparent, non-crystalline and neutral.

4.6 Environmental test

4.6.1 Temperature test 1. Storage temperature range of slide specimen. -40 °C ~ 60 °C. After the test, check the staining and sealing of the slide specimen.

Meets all requirements of 4.4 and 4.5.

4.6.2 Temperature test 2. Maximum temperature for transporting slide specimens. 40 ° C. After the test, check the appearance and cover of the slide specimen, which should comply with 4.2.2 and all requirements of 4.5.

5 Test method

5.1 The test methods for slides and coverslips adopt the provisions of JB/T 8230.3 and JB/T 8230.4.

5.2 The temperature test 1 adopts the method of 4.1 in JY 0213-1994, in which the lower limit temperature of the working conditions is changed to -10 ° C.

Place the cover glass in the slide box with the cover glass facing upward.

5.3 The temperature test 2 adopts the following method. The slide specimen is placed in a slide box vertically and horizontally. The box can be cushioned with a sponge, and the cover is fastened.

The specimen was slowly warmed from room temperature to 40 ° C in the incubator for 4 hours, and it was shaken as it began to warm up. Frequency. 5Hz ~ 55Hz ~ 5Hz;

Amplitude. 0.1mm; vibration direction. vertical. After 4 hours, turn off the power of the incubator and shake it to allow it to naturally drop to room temperature for inspection.

5.4 Specimen selection, staining, and microstructure are examined using a microscope with a magnification of 40 to 1000 times. The specific magnification is in the product standard.
