Scope
GB/T 40458-2021 is the English-translated version of 用于病原微生物高通量检测的核酸提取技术规范.
GB/T 40458-2021 sets the technical requirements for extracting nucleic acids from samples to be screened for pathogenic microorganisms by high-throughput, second-generation sequencing, and describes the matching extraction methods.
It applies to extraction from plant-pathogenic bacteria, fungi and viruses, from vector-borne bacteria, and from plants and plant products. The document explains that extraction has two steps: lysis of the biological sample to free the nucleic acid, and purification to separate it from proteins, polysaccharides, lipids and other cell components.
It calls for the primary structure of the nucleic acid to stay intact, for other molecules and organic solvents to be excluded, and for contamination from dust, aerosols or RNase to be prevented.
It lists the abbreviations DNA, OD, PCR and RNA. It was issued on 20 August 2021 and has applied since 1 March 2022.

5 General requirements
Nucleic acid extraction mainly includes two major steps. lysis and purification of biological samples. Lysis is to free the nucleic acid in the sample in the lysis system In the process, purification is to completely separate nucleic acid from other components in the lysis system, such as proteins, polysaccharides, lipids and other tissues or cell components. The process of leaving.
Nucleic acid extraction should ensure the integrity of the primary structure of nucleic acid. Exclude other molecules, such as proteins, polysaccharides, lipids, organic solvents, etc. To ensure the smooth progress of downstream high-throughput detection tests.
During the test, the external environment (such as dust, aerosol, RNase, etc.) should be prevented from contaminating the nucleic acid extraction, as well as the pathogen itself and the test.
