GB/T 38482-2021 is the Chinese national method for determining animal type I collagen by polyacrylamide gel electrophoresis. It applies to the determination of the type I collagen components of animal-derived collagen products, the check that tells a real collagen from a hydrolysed substitute.

1 Scope
This standard specifies a method for determining the composition of animal-derived type I collagen by polyacrylamide gel electrophoresis (SDS-PAGE).
This standard applies to the determination of type I collagen components in animal-derived collagen products.

3 Terms and Definitions
The following terms and definitions apply to this document.
3.1 Collagen
Three peptide chains of left-handed helical repeating Gly-XY sequence prepared from purified (non-crosslinked) type I collagen Structural protein coiled into a right-handed supercoiled structure characteristic.
Note: Gly is glycine, X is usually proline, and Y is usually hydroxyproline or hydroxylysine.
3.2 TypeIcollagen
Collagen with a complete triple helix structure composed of 2 alpha1 chains and 1 alpha2 peptide chain.
4 Principles
The use of type I collagen has a triple helix structure that other proteins do not have, and with the action of specific collagenase, through Coomassie Brilliant blue was used to determine the staining limit of bovine serum albumin (BSA), and the composition of type I collagen was determined by SDS-PAGE.
5 Reagents or materials
Unless otherwise specified, only analytical grade reagents were used.
5.1 Bacterial collagenase.
5.2 Bovine serum albumin (BSA) standard.
5.4 20mmol/L sodium dihydrogen phosphate solution, pH7.4. END. Draft Version (GB/T 38482-2021).
5.3 Water
It should meet the first-class water regulations in GB/T 6682.
