Scope
GB/T 36821-2018 is the English-translated version of 花生黑腐病菌检疫鉴定方法.
GB/T 36821-2018 is the Chinese national method for detecting and identifying Calonectria ilicicola, the fungus that causes black rot of peanut and red crown rot of soybean.
It gives the pathogen's names and classification, including the asexual stage Cylindrocladium parasiticum, and describes the disease: soil- and seed-borne, spreading fast, blackening cotyledons and then rotting roots and stems.
The fungus grows between 10 °C and 30 °C, best at 25 °C to 28 °C. Identification rests on symptoms on the host, morphological characteristics and PCR-specific amplification, with sampling under SN/T 2122-2015 and a comparison with similar diseases in Annex A. It applies to host plants, plant products and soils such as peanut and soybean, was issued on 17 September 2018 and has applied since 1 April 2019.

3 Basic information and taxonomic status of peanut black rot
English name of the disease. Black rot of peanut, Red crown rot of soybean
Sexual stage scientific name. Calonectria ilicicola Boedijn
Calonectria ilicicola belongs to Ascomycota, Ascomycetes, Sordariomycetidae, Hypocreales, Nectriacea, Calonectria.
Asexual stage scientific name. Cylindrocladium parasiticum Crous, Wingfield
Cylindrocladium parasiticum belongs to the mitosporic fungi (Mitosporic fungi), Hyphomycetes, Hyphomycetales, Hyphomycetaceae, Cylindrocladium.
Peanut black rot is a typical soil and seed-borne disease with serious damage, rapid spread and difficult to control. The pathogens are widely distributed in the soil. In addition to causing legumes such as peanuts and soybeans, they can also cause leaf spots of plants such as tea trees and oil trees. The pathogen first infects the cotyledons to make them black and rot, which in turn infects the roots and stems of the seedlings. In a humid environment, the disease minister has many molds covering the stem base, stems and leaves lost water and wilted death.
It is often confused with peanut new red-shell rot in the field (Table A.1). Peanut black rot bacteria can grow in the range of 10 °C ~ 30 °C, and the optimum growth temperature is 25 °C ~ 28 °C. See Appendix A for additional information on the distribution of pathogens in the world, host plants, symptoms and comparison with similar diseases.
4 Principle of the method
According to the symptoms, morphological characteristics and PCR-specific amplification of peanut black rot bacteria on the host, the detection and identification were carried out.
