Scope
GB/T 30987-2020 is the English-translated version of 植物中游离氨基酸的测定.
GB/T 30987-2020 gives three methods for measuring free amino acids in plants: the automatic amino acid analyser method, high performance liquid chromatography and liquid chromatography-tandem mass spectrometry.
In the first, the free amino acids are extracted with boiling water, separated on the sulfonic acid cation exchange column of the analyser, reacted with ninhydrin at 135 °C and read at 570 nm and 440 nm, identified by retention time and quantified against an external standard curve.
It covers 21 free amino acids, among them glutamic acid, glycine, histidine, lysine, proline, theanine and valine, in tea, Chinese herbal medicines, tobacco leaves and other plant samples.
Annexes give the compounds and their quantitation limits, chromatograms of the mixed standard working solution and reference conditions for the mass spectrometer.
It replaces GB/T 30987-2014, and was issued and has applied since 31 March 2020.

Foreword
This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
This standard replaces GB/T 30987-2014 "Determination of Free Amino Acids in Plants".
Compared with GB/T 30987-2014, the main technical changes are as follows.
--- Added liquid chromatography-tandem mass spectrometry (see Chapter 6);
--- Modified the method of sensitivity expression (see 4.5.5.6, 5.5.6.4, 2014 edition 4.10, 5.10).
1 Scope
This standard specifies the method for determining the content of free amino acids in plants, including the automatic amino acid analyzer method (first method), high-performance liquid phase Chromatograph method (second method) and liquid chromatography-tandem mass spectrometry (third method) three methods.
This standard applies to 21 free amino acids (alanine, arginine, asparagine, asparagus) in tea, Chinese herbal medicines, tobacco leaves and other plant samples Leucine, cystine, gamma-aminobutyric acid, glutamine, glutamic acid, glycine, histidine, isoleucine, leucine, lysine, methionine, phenylpropyl Determination of amino acid, proline, serine, theanine, threonine, tyrosine, valine).

3 Terms and definitions
The following terms and definitions apply to this document.
3.1 Free amino acids
The plant is not combined with other chemical substances, exists in the form of a single molecule or ion, and can be directly leached out of an aqueous solution or other solvent Of amino acids.
4 Fully automatic amino acid analyzer method (first method)
4.1 Principle
After the free amino acid in the sample is extracted by boiling water, it is separated by the sulfonic acid cation exchange column of the amino acid analyzer, and then added at 135 °C Heat, mixed reaction of amino acids and ninhydrin, primary amine and ninhydrin produce blue-purple compounds, secondary amine and ninhydrin produce yellow compounds, respectively in The two derivative products are detected by visible light spectrophotometer at 570nm and 440nm wavelengths, the retention time is qualitative, and the external standard working curve Method quantitative.
4.2 Reagents or materials
Unless otherwise specified, the reagents used in this method are of superior grade. Water is the first-grade water in accordance with GB/T 6682.
4.2.1 Lithium citrate (tetrahydrate).
4.2.2 Lithium chloride.
4.2.3 Citric acid.
4.2.4 Lithium hydroxide.
4.2.5 Sodium borohydride.
