GB/T 28078-2011 sets the quarantine and identification method for Xanthomonas oryzae pv. oryzae (Xoo) and for the rice bacterial leaf spot pathogen Xanthomonas oryzae pv. oryzicola (Xcola) in rice seed and other materials, resting it on plant morphological characteristics, physiological and biochemical characteristics, molecular biology and enzyme immunoassay techniques, and prescribing field observation, isolation and identification, and sample preservation.
Clause 1 applies it to the detection of the two pathogens in rice, in related material and in the environment, and names no exclusion. Clause 2 cites GB 15569-1995 and the ISTA international seed testing protocols.
Clause 3 states the principle of the method, which combines field observation of the morphological characteristics of the rice plant with isolation and culture, molecular biology and enzyme-linked immunological screening, physiological and biochemical identification, and a determination of pathogenicity.
Clause 4 lists the equipment and materials, among them a refrigerated high-speed centrifuge of up to 15 000 r/min, PCR amplification equipment, an incubator at 28 degrees Celsius plus or minus 1 degree, a microscope with 10x to 100x lenses, a balance precise to 0.001 g, an autoclave, a homogenizer running at 4 000 r/min to 8 000 r/min, adjustable pipettes from 0.2 microlitres upwards, sterilized tweezers, scissors and weighing spoons, 90 mm dishes of glass or disposable plastic and 100 mL flasks.
Clause 5 opens the media and reagents and refers the first of them, SPA medium, to A.1, which shows that an annex A holds the media recipes, and the draft breaks off at that point, so the identification provisions themselves cannot be described.

3 PRINCIPLE OF THE METHOD
