Foreword

This document is in accordance with the provisions of GB/T 1.1-2020 "Guidelines for standardization work Part 1: Structure and drafting rules for standardization documents" Drafting.

This document replaces GB/T 21044-2007 "Chinese soft-shelled turtle". Compared with GB/T 21044-2007, except for structural adjustments and editorial changes, In addition, the main technical changes are as follows.

a) The description of external morphological features has been changed (see 5.1.1~5.1.5, 5.1.1~5.1.5 of the 2007 edition);

b) Added the schematic diagram of the appearance measurement of Chinese soft-shell turtle (see 5.2.1);

c) The scalability characteristics have been changed (see 5.2, 5.2 of the 2007 edition);

d) Added the carapace and plastron plates of Chinese soft-shell turtle (see 5.3.2);

e) replace “reproductive characteristics” with “growth and reproduction” (see Chapter 6, Chapter 6 of the 2007 edition);

f) Changed the karyotype diagram, karyotype formula and arm number of Chinese soft-shell turtle (see 7.1, 7.1 of the 2007 edition);

g) The measured graph and scan graph of lactate dehydrogenase (LDH) isozymes in the muscles of Chinese soft-shell turtle were modified (see 7.2, 7.2 of the 2007 edition);

h) Added molecular genetic characteristics (see 7.3);

i) Added the method for determining the age of Chinese soft-shell turtle (see 8.3.2);

j) Changed the pretreatment method for chromosome preparation (see 8.4.1, 8.2.1 of the 2007 edition);

k) The electrophoretic separation method of isozymes has been changed (see 8.5.6, 8.3.2 of the 2007 edition);

l) Added molecular genetic characterization methods (see 8.6);

m) Changed the inspection rules and result determination rules to determination rules (see Chapter 9, Chapter 9 of the 2007 edition);

n) Added the preparation methods of various isoenzyme reagents (see Appendix A).

1 Scope

The main morphological and structural characteristics, growth and reproduction, cytogenetic characteristics, biochemical genetic characteristics and molecular genetics of Chinese soft-shell turtle germplasm were determined.

The paper describes the chemical characteristics of the product, describes the corresponding detection methods, and stipulates the judgment rules.

This document applies to the germplasm detection and identification of Chinese soft-shelled turtle.
