4 Principles
4.1 Antibody Detection Principle Based on immunological principles, PVM antigen was used to detect PVM antibodies in the serum of mice, rats, hamsters, and guinea pigs.
4.2 Principle of Nucleic Acid Detection Based on molecular biology principles, the unique genomic nucleic acid sequence of mouse pneumonia virus can be amplified using nucleic acid amplification methods such as polymerase chain reaction. The technology identifies the target gene or fluorescent signal, thereby obtaining the detection target.
5.1.1 Enzyme-linked immunosorbent assay (ELISA) antigen
5.1.1.1 Specific antigens PVM-infected mice until disease develop, lungs are harvested, ground, and prepared into a 10% suspension. After centrifugation at 3000 rpm for 10 minutes, the supernatant is collected for further infection. BHK21 cells were adsorbed for 1.5-2 hours, then cultured in maintenance medium for 10-14 days. Harvesting was performed when cytopathic effects were observed. The cells were then subjected to three freeze-thaw cycles or more. After acoustic treatment, cell debris is removed by low-speed centrifugation, and the supernatant is then concentrated by ultracentrifugation to prepare ELISA antigen.
