1 Scope

YY/T 1239 is the product standard for the pre-poured agar plate, the ready-to-use culture plate that has largely replaced media prepared in the laboratory. It covers the plate as a manufactured product, which shifts the burden from the technologist to the supplier: fill volume, gel depth and firmness, sterility, moisture, shelf life and the growth performance that a plate has to still deliver at the end of it. Those properties determine whether an organism grows at all, and a laboratory that no longer pours its own media has no way to check them except by trusting the specification. For a manufacturer or a distributor supplying prepared culture plates to Chinese laboratories, this is the governing product standard.

This standard specifies the terms and definitions agar medium, requirements, test methods, marking, labeling, packaging, transportation and storage. This standard applies to conventional microbiological laboratory tests using various solid agar media products, including basic nutrient medium, select Of the medium, the medium identification, susceptibility testing agar medium (Mueler-Hintonagar, MH medium) and the like. This standard does not apply to the requirements of dry powder media.

3 Terms and Definitions

The following terms and definitions apply to this document.

3.1 Medium culturemedium Formulated by the artificially made specifically for microbial culture, separation, and other identification, delivery and preservation of mixed nutrient products.

3.2 Quality control strains qualitycontrolstrain For quality control and performance verification, stable genetic characteristics of microorganisms.

3.3 Colony forming units colonyformingunit; CFU By a single bacteria on solid media growth and reproduction of the formed colonies as a viable count of the number of units.

4 Requirements

4.1 Appearance Medium should be homogeneous, wet, no drops, bubbles; uniform thickness; smooth surface, no protrusions, dents or cracks.

4.2 Thickness Agar We shall meet the following requirements.

a) General agar plate thickness shall not be less than 3mm;

b) MH medium thickness shall be within the range of 4mm ± 0.5mm.

4.3 pH PH of the medium should conform to the specified range.
