Scope
YY/T 0506.7-2014 is the English-translated version of 病人、医护人员和器械用手术单、手术衣和洁净服 第7部分:洁净度-微生物试验方法.
YY/T 0506.7-2014 is the Chinese pharmaceutical industry standard that gives the test method for determining the microbial cleanliness of surgical drapes, surgical gowns and clean air suits.
It is the seventh part of the YY/T 0506 series and sets out the sampling of the product, the extraction of the micro-organisms from it, the culture and the counting of the colonies and the expression of the result as colony forming units per unit area, so that the microbial cleanliness required by the second part of the series can be verified. Issued on 17 June 2014, it has applied since 1 July 2015.

Foreword
YY/T 0506 "Surgical drapes, gowns and clean air suits for patients, clinical staff and equipment" consists of the following parts.
- Part 1: General requirements for manufacturers, processors and products;
- Part 2: Performance requirements and performance levels;
- Part 3: Test methods;
- Part 4: Test method for linting in the dry state;
- Part 5: Test method for resistance to dry microbial penetration;
- Part 6: Test method to determine the resistance to wet bacterial penetration;
- Part 7: Test methods for determination of cleanliness-microbial. This Part is Part 7 of YY/T 0506.
This Part is drafted in accordance with the rules given in GB/T1.1-2009.
1 Scope
The method given in this Part of YY/T 0506 is applicable to the evaluation of cleanliness-microbial of surgical drapes, gowns and clean air suits.
2 Normative references
The following referenced documents are indispensable for the application of this document. For dated references, only the edition cited applies. For undated references, the latest edition of the referenced document (including any amendments) applies.

3 Terms and definitions
For the purpose of this document, the terms and definitions defined in GB/T 19973.1, YY/T 0506.1, YY/T 0506.2 and YY/T 0506.3 apply.
4 Principle
SET the suitable elution procedure, USE the bag peristalsis method to elute the microbials naturally contaminated on the sample, FILTER the eluent by membrane, CULTURE and COUNT the filter membrane, to obtain the total number of microbials eluted from the sample.
5 Instruments and reagents
5.2 Homogenizer, with sterile homogenate bag.
5.3 Filter device, with sterile filter bowl.
5.4 Sterile microporous membrane, with a nominal pore size of 0.45 µm.
5.9 Bacillus atrophaeus ATCC9372
5.10 Nutrient agar medium plate
5.11 dm² template
5.11 Rose red sodium agar medium plate
5.12 Sodium chloride injection
6 Test methods
6.1 Determination of the number of colonies eluted
6.1.1 Sample preparation
On the clean workbench, the test product is folded into five layers in an aseptic manner, and the samples are cut with a sterile 1 dm² template. The uppermost layer and the lowermost layer are used as the protective layer, and the middle three layers are taken as the samples, which are respectively denoted as No. 1, No. 2 and No. 3 samples.
6.1.3 Transfer into the medium
The filter apparatus is placed on a clean workbench, and the filter membrane and filter bowl are loaded onto the filter apparatus in an aseptic manner. Each portion of eluate with a volume of 200 mL is filtered through two filter membranes, 100 mL for each. The surface of the filter membrane with bacteria is placed face up on a nutrient agar medium plate and a rose red sodium agar medium plate, respectively.
6.2 Determination of correction factor
6.2.1 Preparation of inoculum suspension
The Bacillus atrophaeus ATCC9372 spore suspension is prepared by a suitable method and preserved for use. The preserved spore suspension is diluted in gradient with sodium chloride injection to determine the live spore concentration before use, and then use sodium chloride injection to prepare the inoculation suspension, so as to make it contain about 100 CFU of Bacillus atrophaeus spores per 0.1 mL of inoculation suspension. Accurately determine the number of spores in 0.1 mL of inoculation suspension.
6.2.4 Product elution
OPERATE according to 6.1.2.
6.2.5 Transfer into the medium
The filter apparatus is placed on a clean workbench, and the filter membrane and filter bowl are loaded onto the filter apparatus in an aseptic manner. 3 portions of eluate with a volume of 200 mL are filtered through the filter membrane respectively. The surface of the filter membrane with bacteria is placed face up on nutrient agar medium plates.
6.2.6 Cultivation and counting
The three plates affixed with filter membranes are placed in a constant temperature incubator at 35 °C for 3 days. The colony growth is observed every day, and the number of colonies is counted.
6.3 Calculation of cleanliness-microbial
The calculation of cleanliness-microbial is performed according to formula (6).
7 Method description
7.1 Some products may not achieve high recovery rate when eluted using the elution parameters recommended in 6.1.2. If the recovery rate is found to be less than 50 %, adjustments to the elution parameters and/or addition of surfactants may be considered.
7.3 In the routine inspection of a specific product, under the premise that the test method is not changed, the results may be calculated using the obtained correction factor, and it is not necessary to determine the correction factor for each test.
8 Test report
The test report should contain at least the following information. […]
