4 DUS test technical requirements
4.1 Test materials
4.1.1 The applicant for variety rights shall submit test materials that meet the specified quantity and quality requirements at the specified time and place. From a non-testing country or for materials submitted by the region, the applicant shall provide relevant documents such as customs and plant quarantine in accordance with relevant regulations on entry, exit and transportation.
4.1.2 The test materials submitted should be scions or vegetatively propagated plants that have grown for at least 1 year.
4.1.3 The number of submitted test materials shall not be less than 8 or at least shall meet the expansion of 8 seedlings.
4.1.4 The test materials submitted should be plants or scions that are free of pests and diseases and grow normally.
4.1.5 Except for the processing of materials permitted or required by the examination and approval agency, the submitted test materials shall not be subjected to any additional tests that affect the expression of traits.
deal with. If it has been processed, the variety right applicant should provide detailed information on the processing.
4.2 Test method
4.2.1 Test cycle and time
In the case of meeting the test conditions, at least two independent growth cycles are tested.
4.2.2 Test location
The testing of the varieties to be tested shall be carried out in the testing bases and laboratories designated by the approval agency.
4.2.3 Test conditions
The test should be carried out under the condition that the relevant characteristics of the test material can be fully expressed, and the selected test material should at least be planted in the test site or grafting for 2 years.
4.2.4 Test design
4.2.4.1 The variety to be tested should be planted in the same location and environmental conditions as the standard variety and similar varieties. Each variety should be tested with 5 individual plants.
As the basis.
4.2.4.2 If certain parts of the plant need to be extracted as samples for the test, the sample collection shall not affect the observation of the entire growth cycle of the tested plant.
Measurement.
4.2.4.3 Unless otherwise stated, all observations should be made on 5 plants or materials taken from the same part of 5 plants.
4.2.5 Test methods for similar traits
4.2.5.1 Plants (Character No. 2~3 in Table A.1 of Appendix A)
Observe the traits of the plants before leaf development or after dormancy in autumn.
4.2.5.2 Annual branches (character numbers 4~5 in Appendix A Table A.1)
After the branches are lignified in summer to before deciduous, select the current-year branches (two branches per plant) on the sun-facing side of the middle and upper crown of the test plants.
It is a test material for 1-year-old branch characteristics.
4.2.5.3 Leaves (character numbers 6 to 11 in Table A.1 in Appendix A)
After the leaves have matured in summer, select the mid-section leaves of the current-year-old branches in the middle and upper part of the crown of the test plant (choose two branches for each plant, Choose 2-3 leaves for each branch) as the test material for leaf characteristics.
4.2.5.4 Flower buds (character number 12~15 in Table A.1 of Appendix A)
When the flowers are about to bloom, select the flower buds (choose 2 flower buds for each plant) of the branches on the sun-facing side of the crown of the test plant as the flower characteristics Levy test materials.
4.2.5.5 Flowers (Character numbers 16-21 in Table A.1 of Appendix A)
In the blooming period, select the flowers of the branches on the sun-facing side of the top of the crown of the test plant (choose 2 flowers for each plant) as the test of flower characteristics material.
4.2.5.6 Fruit (Character No. 22~27 in Table A.1 of Appendix A)
Before the fruit stops expanding and matures and cracks, select the test plant to produce branches in the upper part of the canopy on the sun side (choose 2 fruits for each plant) as a test material for fruit characteristics.
4.2.5.7 Endocarp (Character No. 28~40 in Table A.1 of Appendix A)
After the fruits are naturally cracked, select the middle and upper fruiting branches of the test plants on the sunny side of the canopy (choose 2 fruits for each plant) as the endocarp special Levy test materials.
4.2.5.8 Nuts (Character No. 41~43 in Table A.1 of Appendix A)
Select the middle and upper fruiting branches of the test plant on the sunny side of the canopy (choose 2 fruits for each plant) as the test material for the characteristics of the nuts.
Observe after drying.
4.2.6 Test methods for individual traits
4.2.6.1 Plant. Growth vigor (No. 1 in Appendix A, Table A.1)
During the dormant period, observe the plant height, crown diameter and new shoot growth, combined with comprehensive evaluation of tree age.
4.2.6.2 Early flowering period (No. 44 in Appendix A, Table A.1)
When 5% of the whole tree blooms, it is recorded as the first flowering period.
4.2.6.3 Relative timing of leaf development and flowering (No. 45 in Appendix A, Table A.1)
In the initial flowering period, select the sun-facing branches in the middle and upper part of the crown of the test plant (choose 2 branches for each plant) as the opposite of leaf development and flowering Time series test materials.
4.2.6.4 Fruit harvest period (No. 46 in Appendix A Table A.1)
When more than 75% of the fruit of the whole tree is mature, it is recorded as the fruit harvest period.
4.2.7 Additional testing
If necessary, the traits in Table A.1 or traits not listed in this guide can be used for additional testing.
5 Evaluation of specificity, consistency and stability
5.1 Specificity
If the difference in traits is constant and significant, it is considered specific.
5.1.1 Constant difference
If the difference between the new variety to be tested and similar varieties is very clear, only one growth cycle is required. In some cases due to environmental factors If the difference between the new variety to be tested and the similar variety is unclear due to the influence of the element, at least two or more growth cycles are required.
5.1.2 Significant difference
Specificity evaluation of quality traits. As long as there is a difference in one trait between the tested variety and similar varieties, it can be determined that the variety has specificity.
Specific evaluation of quantitative traits. if there are at least two discontinuous codes between the tested variety and similar varieties, it can be determined that the variety has Specificity.
Evaluation of the specificity of false quality traits. there is a significant difference in at least one trait between the tested variety and the similar variety, or two of the same trait the difference of discontinuous codes can determine that the variety has specificity.
5.2 Consistency
The consistency judgment adopts the heterogeneous plant method. According to the 1% population standard and 95% reliability probability, no heterotypic plants are allowed among the 5 observed plants.
5.3 Stability
5.3.1 The applied variety meets the consistency requirements in the test, and the variety can be considered as stable.
5.3.2 In special circumstances or when there are doubts, it is necessary to pass another growth cycle test, or the applicant shall provide new test materials to test Does it express the same characteristics as the previously provided test materials?
6 Variety grouping
6.1 Variety group description
Determine the grouping situation of the varieties to be tested according to the grouping traits, and select similar varieties to be included in the specific planting test.
6.2 Grouping traits
6.2.1 Plants. Flower bud distribution (for details, see the character feature number 3 in Table A.1).
6.2.2 Fruit. size (see the character feature number 22 in Table A.1 for details).
6.2.3 Endocarp. Anti-extrusion degree (see serial number 40 in Table A.1 for details).
