GB/T 46144-2025 is the English-translated version of 化学品 鱼类细胞系急性毒性 虹鳟鳃细胞系(RTgill-W1)试验.
GB/T 46144-2025 is the Chinese national standard covering acute fish toxicity measured on gill cells in a dish instead of on live fish — the RTgill-W1 line and the three viability endpoints, the exposure in a sealed plate that stops volatile substances escaping, and the correlation back to the fish LC50. It exists to replace the acute fish test, which kills animals for a number this assay can often give. First edition, in force from 1 December 2025. Issued on 29 August 2025, it has been in force since 1 December 2025.

1 Scope
This document describes the test method for the acute toxicity of chemicals to fish cell lines using the rainbow trout gill cell line (RTgil-W1).
This document is applicable to the testing and evaluation of the acute toxicity of chemicals to fish cell lines.

3 Terms, definitions, and abbreviations
3.1 Terms and Definitions
The following terms and definitions apply to this document.
3.1.1 Cell viability
The percentage of cells that are in a healthy state (i.e., metabolically active, with intact cell membranes and organelles).
3.1.2 effect concentration;ECx
The concentration of test substance that causes a change of x% in cell viability compared to the negative control or solvent control during a given test period.
3.1.3
Leibovitz L-15 medium is supplemented with 5% fetal bovine serum and optional antibiotics.
Note. Commonly used antibiotics include 0.5% gentamicin.
3.1.4 L-15/ex medium
Protein-free medium containing the same salts, galactose, and pyruvate as Leibovitz L-15 medium.
Note. Used for cell exposure experiments.
