This document describes an ion chromatographic method for the determination of Tremella polysaccharide content in Tremella fuciformis.

The following terms and definitions apply to this document.
3.1 Polysaccharides in white jully fungus
Extracted from the basidiomycete Tremella, the monosaccharides include fucose, galactose, glucose, xylose, mannose, galacturonic acid and glucuronic acid.
A polymer compound formed by the dehydration condensation of units with glycosidic bonds.

4 Principle
The sample was extracted with water and the Tremella polysaccharide mixture was hydrolyzed with trifluoroacetic acid. The products were separated by anion chromatography column and equipped with a pulsed amperometric The content of Tremella polysaccharide in the sample was calculated based on the peak area of each monosaccharide by using the ion chromatograph of the detector for determination and the external standard method for qualitative and quantitative analysis.
5 Reagents and Materials
Unless otherwise stated, all reagents used in this method were of analytical grade.
5.1 Water. GB/T 6682, Grade 1. 5.2 Nitrogen (N2). 99.5%.
5.3 Methanol (CH3OH). chromatographic grade.
5.4 Trifluoroacetic acid (C2HF3O2).
5.5 Sodium hydroxide (NaOH). chromatographic grade.
5.6 Sodium acetate (CH3COONa). chromatographic grade.
5.7 Fucose standard (C6H12O5, CAS No.. 2438-80-4). purity not less than 98%, or equivalent standard substance/standard sample.
5.8 Galactose standard (C6H12O6, CAS No.. 59-23-4). purity not less than 98%, or equivalent standard substance/standard sample.
