Scope

GB/T 40187-2021 is the English-translated version of 核酸适配体亲和性和特异性评价技术导则.

GB/T 40187-2021 gives technical guidance for evaluating how strongly and how selectively nucleic acid aptamers bind to their targets. It sets out evaluation rules and determination methods for affinity and specificity, for targets at different levels: metal ions, small molecules, macromolecules, cells and particles, tissues and mixed targets.

The document defines affinity, specificity and the equilibrium dissociation constant KD, which expresses affinity: the smaller the KD, the stronger the interaction between aptamer and target.

Specificity is judged by comparing how much aptamer binds to the target and to non-target substances under the same method and concentration. It refers to GB/T 6682 for laboratory water and GB/T 27403 for quality control in food molecular biology testing. It was issued on 21 May 2021 and has applied since 1 December 2021.

Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.

1 Scope

This standard specifies the evaluation rules and determination methods for the binding affinity and specificity of nucleic acid aptamers to the target.

This standard applies to different levels of target objects such as metal ions, small molecules, macromolecules, cells and particles, tissues, and mixed targets.

Evaluation of the affinity and specificity of the nucleic acid aptamer.

2 Normative references

The following documents are indispensable for the application of this document. For dated reference documents, only the dated version applies to this document. For undated reference documents, the latest version (including all amendments) is applicable to this document.

3 Terms and definitions

The following terms and definitions apply to this document.

3.1 Affinity

The gravitation that exists between the specific target and the nucleic acid aptamer is the inherent binding force of the specific target and the nucleic acid aptamer.

Note: The strength of affinity is generally expressed by the equilibrium dissociation constant KD.

3.2 Specificity

Under the same measurement method, the same amount and concentration of the substance, determine the binding number of the nucleic acid aptamer to the target and its non-target

Note: The higher the ratio of binding quantity, the better the specificity.

3.3 Equilibrium dissociation constant

When the nucleic acid aptamer binds to the target to reach the dissociation equilibrium, the product of the power function of the concentration of each product divided by the power function of the reactant concentration.

Note: In most cases, the nucleic acid aptamer and the target are bound 1.1, and the equilibrium dissociation constant of the nucleic acid aptamer can be simplified to the free state in solution The product of the concentration of aptamer and the concentration of the target substance in the free state is divided by the concentration of the two complexes. Is numerically equal to the target substance The concentration (KD) of the target substance when the aptamer binds halfway. The smaller the KD value, the stronger the interaction between the two.

Where: KD --- equilibrium dissociation constant; cA --- the amount and concentration of the nucleic acid aptamer; cT --- the amount and concentration of the target molecular substance; cE --- The amount and concentration of the compound's substance.
