This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
1 Scope
This standard specifies the serological and molecular biological detection methods for quarantine identification of potato M virus.
This standard applies to the quarantine and identification of potato plants, propagation materials and other host plants that may have potato M virus.

3 Potato M virus basic information
Chinese name. Potato M virus Scientific name. PotatovirusM Abbreviation. PVM Synonym. Kartoffel-Rolmosaikvirus, Ksrtoffel-K-virus, Pototainterveinalmosaicvirus, Potatoleaf Rolingmosaicvirus, PotatoMcarlavirus, potatoparacrinklevirus, PotatovirusE, Solanumvirus 11, Solanumvirus7 Classification status. Betaflexiviridae, Carlavirus.
Route of transmission. The virus can be transmitted by mechanical inoculation and non-persistent transmission of aphids, or through seed potatoes, commercial potatoes, tissue culture seedlings, etc. The material is spread over long distances.

4 Principle of the method
The immunogenicity and genomic characteristics of PVM are the basis for the quarantine and identification of this virus. Establishment of enzyme-linked immunosorbent assay based on PVM immunogenicity Attached assay (ELISA) method and dot enzyme-linked immunosorbent assay (dot-ELISA) method; reverse transcription based on genomic features of PVM PCR (RT-PCR) and reverse transcription loop-mediated isothermal amplification (RT-LAMP) detection methods.
5 Instruments, equipment and reagents
5.1 Instrumentation
Microplate reader, PCR instrument, electrophoresis instrument, biological safety cabinet, small centrifuge, desktop refrigerated centrifuge, constant temperature water bath, pH meter, gel imaging System, ice machine, ultra-clean workbench, autoclave, low temperature refrigerator, electronic balance (0.001g), vortex oscillator, constant temperature incubator, etc.
