Scope

GB/T 36833-2018 is the English-translated version of 马铃薯X病毒检疫鉴定方法.

GB/T 36833-2018 sets out how to detect and identify Potato virus X (PVX) in quarantine. It applies to potato plants, propagation material and other host plants that may carry the virus.

It gives the basic information on PVX, including its classification in the Alphaflexiviridae and Potexvirus and its routes of transmission, which in the field is mainly by contact between plants, tools and seed potatoes.

The methods rest on the genome and on the immunogenicity of the virus: RT-PCR, real-time RT-PCR and RT-LAMP on one side, ELISA, immunochromatographic test strips and dot-ELISA on the other, combined to decide whether a sample carries PVX.

It also lists the instruments and reagents and refers to GB 7331 and SN/T 2122 for seed potato quarantine and sampling. It was issued on 17 September 2018 and has applied since 1 April 2019.

Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.

1 Scope

This standard specifies the quarantine and identification methods for potato X virus.

This standard applies to the quarantine and identification of potato plants, propagation materials and other host plants that may have potato X virus.

2 Normative references

The following documents are indispensable for the application of this document. For dated references, only dated versions apply to this document. For undated references, the latest edition (including all amendments) applies to this document.

SN/T 2122 Quarantine sampling method for inbound and outbound plants and plant products

3 Potato X virus basic information

Chinese name. Potato X virus Scientific name. PotatovirusX Abbreviation. PVX Classification status. Alphaflexiviridae, Potexvirus.

Transmission route. under the natural conditions of the field, the mechanical communication is mainly carried out by the contact friction between different parts of the host plant and the host plants.

In the case of gardening or gardening, it can also be spread by means of tools and tools. Long-distance transmission through the transfer of vegetative propagation materials such as seed potatoes to make. There is information that the potato seed can transmit the virus, but it has little significance in production; no other Solanaceae crops have transmitted the disease.

Poisonous information. According to reports, certain insects such as Melanoplusdifferentialis and Green Talis (Tettigonia) Viridissima's chewing mouthpiece mechanically transmits the virus, Cuscutacampestris and Aphis gossypii (Synchyrtiumendobioticum) can also spread the virus.

See Appendix A for additional information on the Potato X virus.

4 Principle of the method

The genomic characteristics and immunogenicity of PVX are the basis for the quarantine and identification of this virus. Establish reverse transcription based on the genomic characteristics of PVX PCR (RT-PCR), real-time fluorescent reverse transcription PCR (Real-time RT-PCR) and reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay Determination method; establishment of enzyme-linked immunosorbent assay (ELISA) method, immunochromatographic test strip detection method and dot enzyme according to PVX immunogenicity Combined immunosorbent assay (dot-ELISA) method; by an effective combination of these methods, the sample is judged whether it has PVX.
