Scope

GB/T 36816-2018 is the English-translated version of 马铃薯Y病毒检疫鉴定方法.

GB/T 36816-2018 is the Chinese national method for the quarantine detection and identification of potato virus Y (PVY).

It gives basic information on the virus - synonyms, classification in Potyviridae, genus Potyvirus, and its spread through seed potatoes and other vegetative propagation material, mechanical inoculation and vectors in a non-persistent manner.

Detection combines several methods: RT-PCR and real-time RT-PCR, ELISA, immunochromatographic strips, immunoelectron microscopy, diagnosis on host plants, and pyrosequencing to assign strain groups.

Used together, they decide whether a sample carries PVY and its main strains. It applies to potato plants, propagation material and other hosts that may carry the virus, with sampling under SN/T 2122 and reference to GB 7331. It was issued on 17 September 2018 and has applied since 1 April 2019.

Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.

1 Scope

This standard specifies the quarantine and identification methods for potato virus Y.

This standard applies to the quarantine and identification of potato plants, propagation materials and other host plants that may have potato virus Y.

2 Normative references

The following documents are indispensable for the application of this document. For dated references, only dated versions apply to this document. For undated references, the latest edition (including all amendments) applies to this document.

3 Potato Y virus basic information

Chinese name. Potato Y virus

Scientific name. Potato virus Y

Abbreviation. PVY

Synonym. Potato severe mosaic virus, Potato acropetal necrosis virus, Solanum virus 2, Tobacco vein-banding virus, Potato leaf drop streak virus, Tobacco veinal necrosis virus, Potato virus 20

Classification status. Potyviridae, Potyvirus.

Transmission route. vegetative propagation materials such as potato seed potatoes are transmitted, mechanical vaccination can transmit drugs, and various mites spread the disease in a non-persistent manner. Long-distance transmission is accomplished through the trade of asexual propagation materials such as seed potatoes. According to the data, the potato seed can transmit the virus, but in the raw There is little significance in production; no reports have been reported on the seeds of other Solanaceae crops. See Appendix A for additional information on potato virus Y.

4 Principle of the method

The genomic characteristics, immunogenicity and symptom characteristics of the PVY are the basis for the quarantine and identification of the virus. According to PVY and the genomic features of the main strains were established by reverse transcription-PCR (RT-PCR) and real-time fluorescent reverse transcription-PCR (Real-time RT-PCR) detection method; based on the immunogenicity of PVY and major strains, an enzyme-linked immunosorbent assay (ELISA) method and an immunochromatographic strip tester were established method and immunoelectron microscopy; based on PVY and the characteristic characteristics of major strains on different host plants, the identification of host diagnosis method; a pyrophosphate sequencing method was used to identify strain groups based on the genome sequence of PVY. Through the effective combination of these methods, determine whether the sample carries PVY and major strains.
