Scope

GB/T 36809-2018 is the English-translated version of 可可丛枝病菌检疫鉴定方法.

GB/T 36809-2018 sets out how plant quarantine laboratories detect and identify the cocoa pathogen Moniliophthora perniciosa.

Identification is based on the symptoms on the host, the morphological and culture characteristics of the fungus and its molecular biology, with background on taxonomy, host range, distribution and symptoms given in an appendix.

The standard lists the instruments, from stereo and biological microscopes to PCR, electrophoresis and gel imaging equipment, and the laboratory items needed.

It also specifies the reagents for DNA extraction and PCR and the culture media, including potato dextrose agar and a selective PDA medium, with reference to SN/T 1538.1 for medium preparation. It was issued on 17 September 2018 and has applied since 1 April 2019.

Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.

1 Scope

This standard specifies the quarantine and identification methods for Moniliophthora perniciosa.

This standard applies to the quarantine and identification of Moniliophthora perniciosa.

2 Normative references

The following documents are indispensable for the application of this document. For dated references, only dated versions apply to this document. For undated references, the latest edition (including all amendments) applies to this document.

3 Principle of the method

The symptoms of the disease on the host and the morphological, culture and molecular biological characteristics of Moniliophthora perniciosa are the main basis of this standard. See Appendix A for background information on the taxonomic status, host range, distribution, and symptoms of the disease.

4 Equipment

4.1 Instrumentation

Stereo microscope, biological microscope, electronic balance (sensitivity 0.001g), autoclave, ultra-clean workbench, biological incubator, PCR instruments, electrophoresis and gel imaging systems.

4.2 Laboratory equipment

Labels, qualitative filter paper, sample bags, storage bags, hand-held magnifiers, tweezers, scalpels, alcohol lamps, inoculation needles, petri dishes (9.0 cm in diameter), beakers, measuring cylinders, flasks, slides, coverslips, adjustable pipettes (adjustable range 2.5µL~1000µL), centrifuge tubes.

5 Reagents and media

5.1 Reagents

Penicillin, streptomycin, absolute ethanol, potato, glucose, agar, 70% ethanol, sterile double distilled water, 2% CTAB extraction buffer, 10% SDS solution, 3mol/L sodium acetate solution, PCR reagent (including Taq polymerase, dNTP, buffer), phenol, chloroform, isopentanol, Tris-HCl, EDTA (ethylenediaminetetraacetic acid), agarose, molecular weight standard, TAE running buffer.

5.2 Medium

Potato dextrose agar medium (PDA): 200 g of peeled potato, 20 g of glucose, 18 g of agar, add water to 1000 mL, autoclave at 121 °C for 20 min.

PDA selective medium: PDA after autoclaving is cooled to 45 °C ~ 50 °C.
