GB/T 36781-2018 is the English-translated version of 瓜类种传病毒检疫鉴定方法.
GB/T 36781-2018 is the Chinese national method for detecting and identifying the viruses that cucurbit crops carry in their seed.
It covers five common seed-transmitted viruses - cucumber green mottle mosaic virus (CGMMV), cucumber mosaic virus (CMV), melon necrotic spot virus (MNSV), squash mosaic virus (SqMV) and zucchini yellow mosaic virus (ZYMV) - and gives molecular identification methods for the family Potyviridae and the genera Tobamovirus and Sobemovirus.
Identification rests on serological and genomic characteristics, using ELISA and RT-PCR, after sampling to SN/T 2122 and growing the seeds in sterilised soil to the three- or four-leaf stage.
It applies to seed of watermelon, melon, cucumber, pumpkin, bottle gourd, squash, bitter gourd and luffa, and is meant for plant quarantine laboratories. It was issued on 17 September 2018 and has applied since 1 April 2019.

Foreword
This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
1 Scope
This standard specifies five common species-transmitted viruses on cucumber crops--cucumber green mottle mosaic virus (Cucumbergreenmottle) Mosaicvirus, CGMMV), Cucumbermosaicvirus (CMV), Melon Necrotic Spot Virus (Melonnec- Roticspotvirus, MNSV), Squash mosaic virus (SqMV) and small zucchini yellow mosaic virus (Zucchiniyellowmosaicvirus, ZYMV) detection method, which specifies the potato virus family (Potyviridae), tobacco mosaic A molecular identification method for the genus Tobamovirus and the southern bean genus Sobemovirus.
This standard applies to watermelon (Citrullus vulgaris), melon (Cucumismelo), cucumber (Cucumissativus), pumpkin (Cu- Curbitamoschata), Lagenariasiceraria, Cucurbitapepo, Cucurbitamaxima, Quarantine and identification of seeds of melon seeds such as Momordicacharantia and Luffacylindrica.

3 Principle
Serological and genomic characteristics are the main basis for quarantine identification. See Appendix A for background information on melon-type virus transmission.
4 Instruments, reagents and consumables
4.1 Instruments and appliances
Microplate reader, plate washer, PCR instrument, gel imaging system, constant temperature water bath, low temperature refrigerator, ordinary refrigerator, centrifuge, electronic balance, electrophoresis Instruments, electrophoresis tanks, micropipettes, mortars, etc.
4.2 Reagents and consumables
See Appendix B for enzyme-linked immunosorbent assay (ELISA) reagents and Appendix C for RT-PCR assay reagents. Various sizes of tips, centrifugation Tube, enzyme plate, PCR tube, etc.
5 Quarantine and identification
5.1 Sampling
Performed in accordance with SN/T 2122.
5.2 Sample preparation
Seeds are sown in sterilized soil. After growing 3 to 4 true leaves, the plants showing symptoms are numbered separately, and the plants without symptoms are grown.
