GB/T 36778-2018 is the English-translated version of 燕麦花叶病毒检疫鉴定方法.
GB/T 36778-2018 is the Chinese national method for detecting and identifying Oat mosaic virus (OMV), a soil-borne Bymovirus of the family Potyviridae. It sets out three detection routes: double antibody sandwich ELISA (DAS-ELISA), reverse transcription PCR (RT-PCR) and immunoelectron microscopy, the last carried out according to SN/T 1840.
The standard gives the basic information on the virus, the principle of the method, and the instruments and reagents needed, from high-speed refrigerated centrifuges, PCR instruments and microplate readers to the transmission electron microscope, with sampling to SN/T 2122 and water to GB/T 6682.
It applies to the quarantine and identification of the virus in soil around Avena plants and in oat seeds. Plant quarantine laboratories use it. It was issued on 17 September 2018 and has applied since 1 April 2019.

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
1 Scope
This standard specifies the serological, molecular biology and immunoelectron microscopy detection and identification methods for oat mosaic virus.
This standard applies to the quarantine and identification of oat mosaic virus on the soil of the Avena plant, seeds and seeds.

4 Principle of the method
Double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), reverse transcription polymerase chain reaction (RT-PCR) and immunoelectron microscopy (IEM) testing is the main basis for the development of this standard.
5 Instruments and main reagents
5.1 Instrumentation
Electronic balance (sensitivity 0.001g), ordinary balance (sensitivity 0.1g), high-speed refrigerated centrifuge, small centrifuge, ultra-low temperature refrigerator (-80 ° C), ice machine, vortex oscillator, magnetic stirrer, autoclave, pH meter, ultra clean bench, PCR instrument, electrophoresis, electrophoresis Slot, gel analysis imaging system, microplate reader, plate washer, transmission electron microscope.
5.2 Main reagent
All reagents are of analytical grade, and the experimental water should meet the relevant provisions of GB/T 6682. DAS-ELISA detection reagents are shown in Appendix B;
