Scope
GB/T 36775-2018 is the English-translated version of 柑橘斑点病菌检疫鉴定方法.
GB/T 36775-2018 sets out the quarantine detection and identification method for Pseudocercospora angolensis, the fungus that causes citrus spot disease. It applies to citrus seedlings and fruits examined in plant quarantine.
Identification rests mainly on the symptoms on the host and on the morphological and culture characteristics of the pathogen, with a pathogenicity test as supporting evidence.
The standard lists the instruments and laboratory equipment, from stereo and biological microscopes to a clean bench and biological incubator, and the reagents and media, including potato dextrose agar and V8 medium and how they are prepared.
Information on the classification, distribution and hosts of the fungus is given in an annex. It was issued on 17 September 2018 and has applied since 1 April 2019.

Foreword
This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
1 Scope
This standard specifies the quarantine and identification methods for citrus spotted bacteria in plant quarantine.
This standard applies to the quarantine and identification of citrus spotted bacteria in citrus seedlings and fruits.
2 Normative references
The following documents are indispensable for the application of this document. For dated references, only dated versions apply to this document. For undated references, the latest edition (including all amendments) applies to this document.

3 Principle of the method
See Appendix A for the classification, distribution and host information of citrus spotted bacteria. This standard uses the pathogen to host the symptoms, morphological characteristics, The culture characteristics were used as the basis for the main quarantine and identification, and the pathogenicity test was used as the basis for the auxiliary quarantine identification of citrus spotted bacteria.
4 Equipment
4.1 Instrumentation
Stereo microscope, biological microscope, ultra-clean workbench, autoclave, electronic balance (sensitivity 0.01g), biological incubator.
4.2 Laboratory equipment
Labels, qualitative filter paper, sample bags, storage bags, hand-held magnifiers, tweezers, scissors, scalpels, alcohol lamps, slides, coverslips, inoculation needles, Petri dishes, beakers, measuring cylinders, flasks, glass rods, micropipettes (with a range of 2.5 µL to 1000 µL) and pH meter.
5 Reagents and media
5.1 Reagents
Anhydrous ethanol, 0.5% sodium hypochlorite (NaOCl) solution, calcium carbonate (CaCO3), sterile double distilled water.
5.2 Medium
Potato Dextrose Agar Medium (PDA). Weigh 200 g of potato, 15 g of glucose, and 15 g of agar. Potato peeled and cut into small After the block, add water and boil for 20min~30min. After filtering the gauze, add 15g agar, dissolve and add 15g of glucose, and make up the deionized water. 1000 mL, the pH of the medium was adjusted to 5.6, and sterilized at 121 °C for 20 min. Commercialized PDA powder can also be used according to the instructions for use. Formulated.
V8 medium (V8A). 100mL V8 vegetable juice, calcium carbonate 0.2g, agar powder 150g, deionized water to make up 1000mL, 121 °C […]
