Scope
GB/T 36752-2018 is the English-translated version of 藜草花叶病毒检疫鉴定方法.
GB/T 36752-2018 sets out the quarantine methods for detecting and identifying sowbane mosaic virus (SoMV), a member of the genus Sobemovirus, in plant seeds, seedlings and plant products that may carry it.
It combines serological and molecular detection: a double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) on plant samples, and RT-PCR and real-time fluorescent RT-PCR based on the genome of the virus, whose results together decide whether a sample carries SoMV.
It gives basic information on the virus, the principle of the method and the instruments and reagents needed, from refrigerated centrifuges and ultra-low temperature refrigerators to the autoclave and pH meter.
Sampling follows SN/T 2122 and laboratory water GB/T 6682. It was issued on 17 September 2018 and has applied since 1 April 2019.

This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
1 Scope
This standard specifies the serological and molecular biological detection methods for quarantine identification of valerian mosaic virus.
This standard applies to the quarantine and identification of plant seeds, seedlings and plant products that may have valerian mosaic virus.

4 Principle of the method
The serological and genomic characteristics of Valerian mosaic virus is the basis for the quarantine and identification of the virus. According to the relationship between SoMV and antibody specific reaction, double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) for plant samples; based on genomic characteristics of SoMV RT-PCR and real-time fluorescent RT-PCR detection methods were established; through the effective combination of these methods, the samples were judged to have SoMV.
5 Instruments and reagents

Small instantaneous centrifuge (maximum speed 7000r/min).
Ordinary refrigerator (4 °C).
Ultra-low temperature refrigerator (-80 °C).
Ice machine (daily ice making capacity is 110kg).
Vortex oscillator (maximum speed is 2800r/min).
Magnetic stirrer (stirring capacity 50mL~2000mL).
Autoclave (maximum pressure is 0.235MPa, adjustable temperature range is 105 °C ~ 135 °C).
