GB/T 35336-2017 is the English-translated version of 苹果皱果类病毒检疫鉴定方法.
GB/T 35336-2017 is the Chinese national method for detecting and identifying apple fruit crinkle viroid in plant material.
It describes the sampling, the preparation of the material, the molecular tests used to detect the viroid and the criteria by which a result is judged positive or negative, together with the conditions and controls the laboratory has to run.
It applies to the detection and identification of the viroid in plant material that may carry it, that is mainly in nursery stock and in consignments crossing a border.
Plant quarantine services and diagnostic laboratories use it. Issued on 29 December 2017, it has applied since 1 July 2018.

Foreword
This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
1 Scope
This standard specifies the method of quarantine and identification of apple fry virus.
This standard applies to the detection and identification of plant material which may have apple wrinkle fruit virus.

3 Apple wrinkled fruit virus basic information
Scientific name .Applefruitcrinkleviroid. Abbreviation .AFCVd.
Taxonomic status Potato spindle-spiked Pospiviroidae, Apscaviroid.

4 Method principle
Viral detection mainly targets its nucleic acid sequence. The main methods are RT-PCR and nucleic acid hybridization.
5 Equipment, main appliances and main reagents
5.1 Equipment
Electronic balance (flu volume 0.001g), ordinary balance (flu volume 0.1g), high-speed refrigerated centrifuge, small instantaneous centrifuge, general refrigerator, super Low temperature refrigerator (-80 °C), ice maker, vortex shaker, magnetic stirrer, autoclave, pH meter, PCR instrument, microwave ovens, electrophoresis instrument, electricity Swimming pool, gel analysis and imaging system, real-time fluorescence PCR instrument, hybrid furnace, plastic film sealing machine, UV cross-linking instrument, dark box, shaker, constant temperature water Bath, tissue crusher and so on.
5.2 Main appliances
Adjustable pipette (2.5 µL, 10 µL, 20 µL,.200 µL, 1000 µL, 5000 µL), RNase-free tip (10 µL, 20 µL, Mu] L, 1000 [mu] L, 5000 [mu] L), RNAse free centrifuge tubes (1.5 mL, 5 mL, 10 mL), mortar, pestle, magnetic separator Reaction tube (200 µL), real-time fluorescence 96-well reaction plate, hybridization flask, hybrid membrane, hybridization bag, X-ray.
5.3 The main reagent
Unless otherwise specified, all reagents are of analytical grade, the experimental water should be consistent with the relevant provisions of GB/T 6682. RT-PCR test See Appendix B for real-time fluorescent RT-PCR reagents; Appendix C for dot-blot hybridization assays.
