Scope
GB/T 31806-2015 is the English-translated version of 马铃薯V病毒检疫鉴定方法.
GB/T 31806-2015 specifies the phytosanitary quarantine and identification of potato virus V. Clause 1 states that it applies to the quarantine and identification of the virus in potato tubers, potato plantlets and other reproductive material.
Clause 3 lists the equipment and the reagents: the equipment includes a microplate reader, a balance, a pH meter, a trace juicer, a PCR instrument and a real-time PCR instrument, electrophoresis apparatus and an electrophoresis tank, a gel imaging system, an isolation greenhouse, a water bath and a refrigerator, together with micropipettes, ELISA plates, mortar tubes, flower pots and sterilized soil, while the reagents are referred to Appendices B to E, one appendix for each of the four detection methods, the enzyme-linked immunosorbent assay, RT-PCR, real-time fluorescent RT-PCR and the biological assay.
Clause 4 covers sampling and sample preparation; the sampling follows SN/T 2122, and for potato tubers the sample is sown in sterilized soil and grown at 25 degrees Celsius with the symptoms observed, the plants that show no symptoms being grouped in tens and numbered, and leaves are collected for all four detection methods.
For tissue culture material, each plant in the culture bottle is sampled and the sampled bottle serves as one sample for the same four methods. The draft published on the page breaks off at the end of that subclause, so the detection methods themselves and the appendices are not shown.
Clause 2 cites SN/T 2122 on the sampling of plants and plant products at entry and exit. The foreword records that the standard is centralized by the National Standardization Technical Committee of Plant Quarantine (SAC/TC271). It was issued on 3 July 2015 and has applied since 27 November 2015.

1 Scope
This standard specifies the phytosanitary quarantine and identification of potato virus V.
This standard applies to quarantine and identification of potato tubers, potato plantlets and other reproductive material V virus.

3 Equipment and reagents
3.1 Equipment
Microplate reader, the balance (a sense of volume 1/10000g), pH meter, trace juicer, PCR, real-time PCR instrument, electrophoresis, electrophoresis tank, Gel imaging system, isolate the greenhouse, water bath, cold refrigerator.
Micropipette (2µL, 10µL, 20µL, 100µL, 200µL, 1000µL), ELISA plate, mortar tubes, flower pots, sterilization Soil.
3.2 Reagents

4 Sampling and Sample Preparation
4.1 Sampling
Sampling conducted in accordance with the provisions of SN/T 2122's.
4.2 Preparation of the samples of potato tubers
Potato tubers sown in sterilized soil, grown and symptoms observed at 25 °C. Grow to be about 10 after showing symptoms of leaf explants Line numbers do not show symptoms of the plant grouping (for a group of 10) and number. Leaves collected for enzyme-linked immunosorbent assay, RT-PCR detection, real Fluorescent RT-PCR, and biological assays.
4.3 Tissue Culture Sample preparation
The tissue culture bottle of each plant were sampled, the sampled tissue culture bottle product as a sample for the ELISA test sample Fixed, by RT-PCR, real-time fluorescence RT-PCR, and biological assays.
