7.1 Extraction

7.1.1 Pure milk, yogurt, fruit milk, cheese and so on

Weigh the sample 5 g (accurate to 0.01 g) in 50 mL centrifuge tube, add 20 mL of acetonitrile and 5 g of anhydrous sodium sulfate, Quality extraction 1 min, adding 2 g sodium chloride, oscillation, centrifugation at 4 000 r/min 3 min. Take the supernatant and remove 10 mL of the residue Acetonitrile repeated extraction 1 times, combined supernatant, 40 o C water bath in the rotary evaporation concentrated to near dry, add 5 mL of methanol dissolved to be purified.

7.1.2 Milk powder and so on

Weigh the sample 3 g (accurate to 0.01 g) in 50 mL centrifuge tube, add 5 mL of water, 30 o C water bath for 10 min. again Add 20 mL of acetonitrile and 5 g of anhydrous sodium sulfate, homogenously extracted 1 min, adding 2 g of sodium chloride, shaking at 4 000 r/min Centrifuge 3 min. The supernatant was taken and the residue was extracted again with 10 mL of acetonitrile. The supernatant was combined and rotated in a 40 o C water bath Evaporation concentrated to near dry, add 3 mL of methanol dissolved to be purified.

7.2 Purification

After activation of ENVI-18 solid phase extraction with 10 mL of methanol, the active solution was discarded and the 2 mL extract was taken. With 15 mL of methanol Perform elution (flow rate no more than 1 mL/min). Collect the sample and the entire eluent in a 100 mL flask and rotate in a 40 o C water bath Evaporate to near dryness. Dry with nitrogen, methanol - water solution (3 2, V/V) dissolved and fixed to 1.0 mL, filter membrane for liquid phase color Determination and confirmation of spectrum - tandem mass spectrometry.

7.3.3 Determination and confirmation of chromatography

According to the sample content of the sample, the selected concentration of similar matrix mixed standard working solution, matrix mixed standard working solution and to be the response values of 14 carbamate pesticides in the sample solution should be within the linear range of the instrument. Matrix mixed standard working solution and sample solution Determination of volume by volume. For more information on the multi-reaction monitoring (MRM) chromatograms of 14 carbamate pesticide standards, see Appendix B.

The standard solution and sample solution are determined according to the conditions specified in 7.3.1 and 7.3.2. If the same retention time as the standard solution in the sample solution is Peak appears, then its confirmation. The results show that the retention time of the chromatographic peaks is consistent with that of the reference material, and after deducting the background the selected ions are present; the abundance of the selected ions is greater than the relative abundance of the ions associated with the standard sample Caused by similarity within the allowable deviation (see Table 2), you can determine the existence of the corresponding sample in the sample. Using the standard curve of the matrix solution Quantitative method.
