B.5.2 Sample preparation B.5.2.1 Preparation of non-wool dyed fabric samples Put the above mixed sample into a 250mL round bottom flask, add 100mL methanol, 20mL 4mol/L hydrochloric acid, 80°C magnetic stirring and heating Reflux for 2h, cool to room temperature, and filter under reduced pressure to obtain the dye extract of the sample. Put it on the vacuum rotary evaporator, at 50°C±1°C Concentrated under low vacuum and spin-dried, and then dried under vacuum at 50°C for 24h to obtain a powdery solid.
Note. The above extraction conditions have been verified to be applicable. During the experiment, the extraction method can also be adjusted according to the actual situation.
B.5.2.2 Preparation of dyed fabric samples containing wool Put the above mixed sample into a 250mL round bottom flask, add 100mL methanol, 10mL 12mol/L concentrated hydrochloric acid, and magnetically stir at 80°C Heat to reflux for 2h, cool to room temperature, and filter under reduced pressure to obtain the dye extract of the sample. Put it on the vacuum rotary evaporator, at 50°C± Concentrate and spin dry at 1°C under low vacuum. Then the dye is separated by column chromatography (the adsorbent is 100-200 mesh silica gel, and the eluent is ethyl acetate and methanol, V ethyl acetate. V methanol=1.0~0.1), the specific operation is as follows.
First put a certain amount of cotton into the bottom of the column, then load 100-200 mesh silica gel and compact it (the height of silica gel is 5 times the diameter of the column), and then Add ethyl acetate solution to the silica gel surface, and the above low vacuum concentration and spin-dried sample dissolved in a small amount of methanol and then added to the column (add it should be noted that the silicone surface should not be washed away). Use a dropper to add a certain amount of ethyl acetate along the inner wall of the column, and then pour the prepared eluent onto the column the ball-shaped funnel is pressurized, and the gradient elution method is adopted. First use an Erlenmeyer flask to take the solvent in the column, when the colored solution in the column reaches a certain level when the height is high, use test tube to collect. Finally, put all the dye solutions collected on a vacuum rotary evaporator at 50°C±1°C under low vacuum concentration.
Dry, and finally vacuum-dried at 50°C for 24h to obtain a powdery solid.
Note. The above extraction conditions have been verified to be applicable, and the extraction method can also be adjusted according to actual conditions during the experiment. In addition, the column chromatography eluents ethyl acetate and methanol the ratio can be adjusted accordingly according to the polarity of the dye.
B.5.3 Test
Accurately weigh 5mg of the powdered solid for elemental analysis and measure its sulfur content.
B.6 Determination of sulfur-containing synthetic dyes If the sulfur content exceeds 0.1%, it is determined that the dyed fabric contains synthetic dyes.
Appendix C
(Normative appendix) Identification of 18 kinds of vegetable dyes C.1 Principle as shown in Appendix A, 18 vegetable dyes have their own dye types, dye recognition pigments and recognition structures. Integrate with Appendix D Atlas, 18 kinds of plant dyes were identified by comparing the retention time of the sample and the standard sample liquid spectrum and the mass-to-nucleus ratio in the mass spectrum.
C.2 Reagents and instruments Methanol, concentrated hydrochloric acid (10mL of concentrated hydrochloric acid is diluted with water to 30mL to obtain 4mol/L hydrochloric acid), reactor (with a closed block, approximately 100mL round bottom flask made of hard glass), vacuum filtration device, vacuum rotary evaporator, liquid-mass spectrometer (with diode Array detector and XCT type ion trap mass detector) etc.
C.3 Test process
C.3.1 Accurately pipette an appropriate amount (about 1.0 mL) of methanol or other suitable solvents into the round bottom flask containing the powdered solid in Appendix B Medium, mix well and let stand.
C.3.2 According to the dye type and identification pigment indicated in the instructions for use, select the appropriate identification pigment standard sample working solution (Appendix D.1.1).
C.3.3 Take an appropriate amount of sample and standard sample into the chromatograph, and use the analysis conditions of the corresponding standard sample in Appendix D for testing. If so the instructions clearly indicate that there are two or more identification pigments, and the sample and the standard sample of each identification pigment should be injected into the chromatograph respectively, in turn test.
Note. Because the liquid-mass spectrogram test results depend on the instrument parameters used, it is impossible for Appendix D to give the general parameters of chromatographic analysis, but the operation in Appendix D is used.
The working conditions proved to be suitable for the test. In the actual test, the instrument parameters can be adjusted according to the instrument used.
C.4 Judgment
C.4.1 If there are obvious peaks in the liquid phase spectrum of the sample, and the retention time and mass-nucleus ratio are not in the liquid phase-mass spectrometry spectrum provided in Appendix D.
Inside the enclosure, it is considered that the fabric contains synthetic dyes or vegetable dyes other than those specified in this standard.
C.4.2 If the peak retention time and mass-nucleus ratio of the sample are consistent with the selected standard sample, and combined with Appendix A Table 1, it is considered that the fabric contains the standard sample identifies the vegetable dye corresponding to the pigment.
Appendix D
(Informative appendix) Liquid phase-mass spectra of 18 vegetable dye identification pigment standards D.1 Test conditions D.1.1 Reagents D.1.1.1 The solvent used in liquid chromatography is chromatographically pure, and the color standards are all analytically pure.
D.1.1.2 Grade 3 water meets the requirements of GB/T 6682.
